摘要 核酸药物主要指具有遗传特性和药理活性的含核苷酸或脱氧核苷酸结构的化合物,是一类重要的药物,可用于肿瘤、组织再生、创面愈合、肺纤维化、炎性疾病、微生物感染等治疗。核酸从体外进入体内并进入细胞内的过程,存在较多障碍,如降解、沉淀、蛋白结合等。纳米载体可携带核酸药物进入细胞内,也可在细胞外释放药物后再被细胞摄取,因此核酸纳米制剂的研究意义重大。本综述全面论述了核酸药物分类、理化性质和入胞过程,以及主要纳米制剂类型、新型递送技术、体内过程及研发前景,旨在为核酸药物新制剂研发提供参考。 Nanocarriers can be the vehicles to deliver nucleic acids into cells or release them outside the cells followed by cell internalization. Therefore,nucleic acid-loaded nanoscale formulations is important. This review summarizes the classification,physical and chemical properties,internalization,nanoscale formulation types,novel delivery techniques,in vivo dynamics,and prospective of nucleic acid. It will provide references for research and development of nucleic acid formulations.
