电针对帕金森病模型大鼠脑黑质内Lamp2a、Hsc70、α-syn表达的影响 被引量:13

Effect of Electroacupuncture on Expression of Lamp2a,Hsc70 and α-syn in Substantia Nigra of Parkinson’s Disease Model Rats Induced by Rotenone

摘要 目的:观察电针对鱼藤酮诱导的帕金森病(Parkinson’s disease,PD)模型大鼠脑黑质内Lamp2a、Hsc70、α-syn表达的影响,探讨电针治疗帕金森病的可能作用机制。方法:雄性SD大鼠随机分为正常组、假手术组、模型组、电针治疗组,每组15只。采取颈背部皮下注射鱼藤酮法制备帕金森病模型,假手术组只注射不含鱼藤酮的等量葵花油乳化液。正常组、假手术组、模型组不进行任何治疗;电针治疗组在造模完成后取穴"风府"、"太冲",连续波,频率120次/min,每次治疗20 min,每天1次,疗程为28 d。免疫组化法检测模型大鼠黑质内TH、α-syn的阳性表达,并用Western blot法检测模型大鼠黑质内Lamp2a、Hsc70、α-syn的表达。结果:模型组大鼠表现出明显的PD症候群特征,电针干预后模型大鼠行为学评分明显降低(P〈0.01);免疫组化法及Westernblot检测显示电针治疗通过上调Lamp2A、Hsc70水平能抑制错误折叠蛋白α-syn的表达,提高TH的阳性表达。结论:电针可以抑制具有细胞毒性作用的α-syn的集聚,保护多巴胺能神经元,抑制帕金森病的发展,其机制可能与干预分子伴侣介导的自噬有关。 Objective: To observe the effect of electroacupuncture on the expressions of Lamp2a,Hsc70 and α-syn in the brain substantia nigra of Parkinson's disease rat model induced by rotenone and explore the mechanism of electroacupuncture on Parkinson's disease. Methods: Male SD rats were randomly divided into normal group,sham-operation group,model group and electroacupuncture( EA) treatment group,15 rats in each group. The PD model was established by subcutaneous injection of rotenone at the back of the neck,and rats in the sham-operation group were injected with sun-flower oil that did not include rotenone. The normal group,sham-operation group and model group were not given any treatment. After the models were established,EA was applied in the treatment group at each treatment 20 min,once a day and the treatment period was 28 days respectively. The expressions of TH and α-syn in the substantia nigra of the rats were detected by immunohistochemistry. The expressions of Lamp2 A,Hsc70 and α-syn in the substantia nigra of the rats were detected by Western blot. Results: Rats in model group showed obvious characteristics of PD syndromes.The behavioral scores significantly decreased after EA intervention( P 〈0. 01). Immunohistochemistry and Western blot analysis showed that electroacupuncture could inhibit the expression of misfolded protein α-syn and increase the expression of TH by up-regulating Lamp2Aa and Hsc70 protein levels. Conclusion: Electroacupuncture can inhibit the accumulation of α-syn with cytotoxicity,protect dopaminergic neurons and inhibit the development of Parkinson's disease. The mechanism may be related to the intervention of molecular chaperone mediated autophagy.
