参草通脉颗粒影响慢性心衰大鼠心肌重构的作用机制研究 被引量:6

摘要 目的:分析参草通脉颗粒对心力衰竭大鼠心肌重构的作用机制。方法:取80只大鼠,予左冠状动脉前降支结扎,同时配合力竭式游泳、节食等方法做成慢性心衰模型,设正常组20只,将成功造模的大鼠分为模型组、中药组、西药组3组,以灌胃给药或蒸馏水28 d。以实时荧光定量PCR技术及SABC免疫组化法分别检测MMP-1、MMP-2、MMP-9蛋白的基因表达情况,及Ⅰ型胶原、Ⅲ型胶原的蛋白表达情况。结果:相对于正常组,其余3组大鼠MMP-1、MMP-2及MMP-9蛋白的基因表达水平显著升高(P〈0.05),Ⅰ型胶原、Ⅲ型胶原的表达中药组较西药组升高明显(P〈0.05)。结论:参草通脉颗粒抑制慢性心衰心肌重构的机制可能与Ⅰ、Ⅲ型胶原和基质蛋白酶的表达有关。 Objective: To analyze the effect of of Shencao Tongmai Granule on myocardial remodeling in rats with heart failure. Methods: Eighty rats,by the left anterior descending coronary artery ligation method with the exhaustive method type swimming and diet,was set up the chronic heart failure model. The successful model rats were divided into model group,TCM group,western medicine group and normal group,giving medicine or distilled water by gavage for 28 days.We detected MMP-1,MMP-2 and MMP-9 protein and gene expressions and the expression of collagen type I and type Ⅲ collagen protein by real-time fluorescent quantitative PCR technique and SABC immunohistochemical method. Results: Compared with the normal group,model group and the other groups' rats MMP-1,MMP-2 and MMP-9 levels increased significantly( P 〈0. 01). The model group's expressions of type I collagen and type Ⅲ collagen were significantly decreased compared to those of the normal group( P 〈0. 01). Compared with the model group,Chinese medicine group and Western medicine group's MMP-2,MMP-1 and MMP-9 significantly decreased( P 〈0. 05). The expressions of collagen type I and type Ⅲ collagen increased significantly( P 〈0. 01). The western medicine group and Chinese medicine group's MMP-1,MMP-2 and MMP-9 contents in myocardial tissue had no significant difference( P 〈0. 05). The Chinese medicine group's expressions of type I collagen and type Ⅲ collagen increased significantly compared with those of the Western medicine group( P 〈0. 05). Conclusion: The relevant expression mechanism of Shencao Tongmai Granule inhibiting chronic heart failure myocardial remodeling may be related with I,Ⅲ collagen and matrix metalloproteinase.
