摘要
目的观察在高糖环境下白念珠菌对人永生化角质形成细胞(Ha Ca T)表达TLR2mRNA的影响。方法分别在高糖和低糖DMEM培养基下共同培养Ha Ca T细胞与白念珠菌菌悬液,分别为高糖(HG)实验组,低糖(LG)实验组。并将两种培养基下未受白念珠菌感染的Ha Ca T细胞做对照,分别为HG对照组,LG对照组。观察Ha Ca T细胞与白念珠菌培养情况,并在第1h、6h、24h通过逆转录聚合酶链反应(RT-PCR)的方法检测4组中TLR2 mRNA表达量的变化。结果在第1h、6h、24h,TLR2 mRNA表达量始终表现为LG实验组高于HG实验组,LG对照组高于HG对照组,且实验组高于对照组(P<0.01)。HG实验组与LG实验组中TLR2mRNA均于第1h开始增多,6h达到高峰,到第24h均有所下降;且HG实验组在第24h与LG实验组相比下降得更明显(P<0.01);而HG对照组与LG对照组则始终无明显改变(P>0.01)。结论 Ha Ca T细胞感染白念珠菌后,高糖环境不利于角质形成细胞TLR2 mRNA的分泌,推测糖尿病患者易患白念珠菌感染性皮肤病与Ha Ca T细胞表达TLR2 mRNA减少有关。
Objective To explore the differential expression of TLR2 mRNA secreted by HaCaT cells counteracting C.albicans under high glucose condition. Methods HaCaT cells were cultured in high-glucose Dulbecco's modified Eagle's medium(DMEM)and low-glucose DMEM respectively.They were divided into four groups: HG-control group, LG-control group, HG-test group and LG-test group.Then, HaCaT cells in HG-test group and LG-test group were exposed to C.albicans hyphae for 1, 6, and 24 hours.The growth of HaCaT cells with C.albicans, and the expression levels of TLR2 mRNA were detected through Real-Time PCR(RT-PCR)at different periods in each group.Results The TLR2 mRNA in the LG groups was higher than that in the HG groups at each time point both in the test and control groups, and the test groups were higher than the control groups both in the LG and HG groups(P 〈0.01).The expression levels of TLR2 mRNA increased and then decreased both in the HG-test group and LG-test group, and the HG-test group declined more sharply than the LG-test group after 24 hours of treatment with C.albicans.(P〈0.01).However, There were no obvious changes in the HG-control group and LG-control group during the 24 hours' observation(P〉0.01).Conclusion High glucose condition is a disadvantage for HaCaT cells to secrete TLR2 mRNA after infected with C.albicans.We deduced that it is related to the susceptibility of C.albicans for keratinocytes to express lower TLR2 mRNA in diabetic patients.
