内质网应激及介导细胞凋亡信号转导研究进展 被引量:25

摘要 近年来,内质网应激(ERS)和它所诱导的细胞凋亡被认为在慢性疾病、神经退行性疾病、癌症和糖尿病等疾病的病理生理中发挥重要作用。当内质网(ER)正常功能紊乱时,会扰乱细胞内环境,引起ERS,从而激活一系列进化保守的自我保护性信号通路统称为未折叠蛋白质反应(UPR)。最初,UPR旨在恢复ER内部平衡,保护细胞存活,但如果这些尝试失败,UPR将转而激活凋亡级联反应,导致细胞凋亡。文章对ERS反应进行介绍,综述了ERS激活跨膜蛋白激酶(IRE1)、蛋白激酶R样内质网激酶(PERK)、活化转录因子6(ATF6)介导的促存活通路及ERS介导的包括增强子结合蛋白同源蛋白(CHOP)、半胱天冬氨酸蛋白12(Caspase12)、c-Jun氨基末端激酶(JNK)等促细胞凋亡途径的可能机制。
