摘要
目的:研究补阳还五汤对脑缺血再灌注模型大鼠血小板聚集及血栓形成的影响。方法:将75只健康SD雄性大鼠随机分为对照组、假手术组、模型组、西药组和中药组各15只,对照组、假手术组大鼠、模型组大鼠给予生理盐水灌胃;西药组大鼠给予氯吡格雷灌胃(7.0 mg·kg^(-1));中药组大鼠给予补阳还五汤灌胃(20 g·kg^(-1))。所有大鼠连续喂养14 d,第15天建立脑缺血再灌注损伤大鼠模型;测定各组大鼠血清血栓素B2(thromboxane B2,TXB_2)和6-酮-前列腺素F1α(6-keto-PGF1α)水平、凝血指标、血小板聚集率、血小板聚集抑制率以及血栓形成时间。结果:正常组与假手术组大鼠各项指标比较,差异无统计学意义(P>0.05);与对照组和假手术组比较,模型组血清TXB_2升高、6-keto-PGF1α降低,比值A明显升高,1 min、2 min、5 min和最大血小板聚集率均明显升高,凝血酶原(prothrombin,PT)、活化部分凝血活酶时间(activated partial thromboplastin time APTT)、凝血酶时间(thrombin time,TT)明显减少,纤维蛋白原(fibrinogen,FIB)增加,血栓形成时间减少,差异具有统计学意义(P<0.05);与模型组比较,西药组和中药组血清TXB_2降低、6-keto-PGF1α升高,比值A明显降低,1 min、2 min、5 min和最大血小板聚集率均明显低于模型组,西药组、中药组血小板聚集抑制率分别为(45.8±4.1)%、(44.3±4.8)%,西药组和中药组PT、APTT、TT明显增加,FIB减少,血栓形成时间增加,差异均有统计学意义(P<0.05)。结论:补阳还五汤可降低脑缺血再灌注损伤大鼠血清TXB_2、血小板聚集率,升高6-keto-PGF1α,且能有效改善血小板聚集和凝血功能,明显增加血栓形成时间,改善缺血再灌注损伤情况,保护脑组织。
bjective: To study the effect of Buyang Huanwu Decoction on platelet aggregation and thrombosis in rats with cerebral ischemia reperfusion. Methods: 75 healthy male SD rats were randomly divided into control group,sham operation group,model group,western medicine group and Chinese medicine group,with 15 rats in each group. Rats in the control group,the sham operation group and the model group were given saline gavage; while the rats in western medicine group were given intragastric administration of clopidogrel(7. 0 mg·kg^-1). The rats of the Chinese medicine group were given Buyang Huanwu Decoction by intragastric administration(20 g·kg^-1). All rats were fed continuously for 14 days and on the fifteenth day the model of cerebral ischemia-reperfusion injury in rats was established. Serum TXB2 and 6-keto-PGF1 alpha content,coagulation parameters,platelet aggregation,inhibition of platelet aggregation and thrombus formation time of all of the groups were examined. Results: There was no significant difference between the normal group and the sham operation group rats in all indexes(P〈 0. 05). Compared with that of the control group and the sham operation group,the western medicine and Chinese medicine group rats' serum TXB2 increased,6-keto-PGF1α decreased the ratio of alpha A increased of and significantly(P〈 0. 05); 1 min,2 min,5 min and the maximum platelet aggregation rate of the western medicine and Chinese medicine group were significantly increased,while PT,APTT,TT of them decreased significantly; FIB increased thrombosis time is reduced,with the differences being statistically significant(P〈 0. 05);compared with that of the model group,western medicine group and Chinese medicine group rats' decreased and serum TXB2,6-keto-PGF1 were elevated,with the ratio of that being significantly(P〈 0. 05). 1 min,2 min,5 min and maximum platelet aggregation rate were significantly lower than those in the model group,western medicine group,Chinese medicine group aggregation rat
