摘要
目的评定LC-MS/MS法测定人血清门冬酰胺浓度的不确定度。方法采用A类评定评价重复测量引入的不确定度,B类评定评价标准品称量、标准溶液和血清标准样品制备、血清样品提取过程、LC-MS/MS法测定及标准曲线拟合引入的不确定度,并计算合成和扩展不确定度。结果当置信概率为95%时,血清低(6μmol·L^(-1))、中(80μmol·L^(-1))、高(150μmol·L^(-1))浓度门冬酰胺的扩展不确定度分别为0.46μmol·L^(-1)、3.52μmol·L^(-1)及6.82μmol·L^(-1)。结论 LC-MS/MS法测定人血清门冬酰胺浓度的不确定度主要由样品提取过程和标准曲线拟合引入。
OBJECTIVE To evaluate the measurement uncertainty for the determination of asparagine in human serum using LCMS/MS. METHODS The uncertainty that caused by repeat measurement was evaluated in terms of type A,while those caused by weighing,stock solution and standard serum preparation,serum sample extraction,LC-MS/MS determination and calibration curve regression were analyzed according to type B. The expanded and combined uncertainties were calculated by all the above components.RESULTS The expanded uncertainties for asparagine at low(6μmol·L^-1),middle(80μmol·L^-1) and high(150 μmol·L^-1)concentrations were 0.46 μmol·L^-1,3.52 μmol·L^-1 and 6.82 μmol·L^-1,respectively(P= 95%). CONCLUSION The measurement uncertainty for determination of asparagine is mainly caused by sample extraction and calibration curve regression.
