摘要
目的研究维生素D在健康对照组以及单纯糖尿病组、单纯重度慢性牙周炎组、Ⅱ型糖尿病伴重度慢性牙周炎组患者外周血中的表达情况。方法根据分组情况,将142名研究对象采用PCR和酶切的方法对外周血中维生素D受体FokⅠ位点等位基因进行检测。比较对照组与实验组间基因型及等位基因携带频率的不同,运用独立样本t检验及方差分析比较携带患病组与健康对照组临床指标的差异。结果维生素D受体FokⅠ位点Ⅱ/Ⅱ纯合子基因型为5例,其携带频率为13.16%,而Ⅱ型等位基因携带频率为36.84%。统计分析结果显示实验组间基因携带频率有统计学意义(χ~2=17.081,P=0.009﹤0.01),等位基因频率分布亦存在统计学意义(χ~2=14.560,P=0.002﹤0.01)。结论Ⅱ型糖尿病伴重度慢性牙周炎患者的易感等位基因为维生素D FokⅠ位点等位基因Ⅱ纯合子,组间比较有统计学意义。
Objective To study the expression of vitamin D in peripheral blood of the control group （ the healthy） , group simply with diabetes, group simply with severe chronic periodontitis, and group of type Ⅱ diabetes with severe chronic periodontitis. Methods According to the grouping, the allele of vitamin D receptor Fok Ⅰ locus in peripheral blood of 142 subjects were examined by PCR and restriction enzyme digestion. The different genotypes and carrying frequency of alleles of the control group and the experimental groups were compared. The difference in clinical indexes between the healthy control group and other groups were compared with in- dependent-sample t-test and variance analysis. Results There were 5 cases of vitamin D receptor Fok I locus Ⅱ / type Ⅱ homozy- gous genotype with carrying frequency of 13.16% , while the carrying frequency of type Ⅱ allele was 36. 84%. The results of statisti- cal analysis showed that there was statistically significant difference in the carrying frequencies of genes in the experimental groups （ χ2 = 17.081, P = 0. 009 ） , and also in the allele frequency distribution （ χ2 = 14. 560, P = 0. 002 ）. Conclusion The susceptible allele of patients with type Ⅱ diabetes mellitus and severe chronic periodontitis are homozygous for the type Ⅱ allele of vitamin D Fok Ⅰ , with statistical significance among the groups.
