摘要
目的研究前列腺素E2受体亚型EP1在不明原因复发性自然流产(URSA)中表达水平变化,分离鉴定子宫平滑肌细胞,并用特异性siRNA抑制EP1的表达,观察其对子宫平滑肌收缩率的影响,为抗RSA药物研究提供新的理论依据。方法用荧光定量PCR检测两组子宫平滑肌细胞膜上EP1的表达分布情况;应用图像分析系统测量正常组和URSA组病人子宫平滑肌细胞收缩率;用特异性siRNA抑制URSA组子宫平滑肌细胞EP1的表达,分析其对在子宫平滑肌细胞收缩率的影响。结果 URSA组子宫平滑肌细胞中EP1的表达水平显著高于正常组(P<0.01),同时URSA组中子宫平滑肌细胞的收缩率显著高于正常组(P<0.01);EP1特异性siRNA和阴性对照siRNA处理URSA组子宫平滑肌细胞后,EP1特异性siRNA处理子宫平滑肌细胞收缩率较阴性对照siRNA处理组显著下降(P<0.05)。结论在RSA病人中,外在刺激或内在因素导致EP1表达上调可能是促使子宫的异常收缩而发生流产的一个重要原因。因而PGE2/EP1系统可作为治疗RSA的一个潜在靶点。
Objective To research the change of the level of EP1,which is the E2-prostanoid receptor,in unexplained recurrent spontaneous abortion,to isolate and identify the cells of uterine smooth muscle,and to suppress the expression of EP1 through siRNA.Methods Realtime fluores-cence quantitative PCR was used to test the distribution of EP1 uterine smooth muscle cytomembrane of two groups.Percentage of contraction of uterine smooth muscle cytomembrance of normal group and URSA group was measured by image analysis system.Specificity siRNA was used to suppress the expression of EP1 of uterine smooth muscle cytomembrance in URSA group to analyze its influence on the percentage of contraction of uterine smooth muscle cytomembrance.Results In total,the level of EP1 was found higher in URSA group than in normal group（P 0.01）,consistently,cell contraction was found significantly higher in RSA group than in control group（P 0.01）.After specificity siRNA and negative control siRNA on uterine smooth muscle cell,EP1 significantly decreased the level of cell contraction in uterine smooth muscle cytomembrance compared with negative control group（P 0.05）.Conclusion Results suggest that upregulation of EP1,which is induced by external stimulus and intrinsic factor,may become an important factor of abortion induced by abnormal contraction of uterine.So PGE2/EP1 system can become a target spot of RSA treatment.
