摘要
提高油菜籽品质一直是育种家们不懈追求的目标之一,不同的栽培措施对油菜不通脂肪酸合成有一定影响,为从分子方面促进油菜脂肪酸组成改良研究的发展,以甘蓝型油菜420为材料,采用三元二次回归正交组合设计方法,对不同生育时期、不同栽培条件下叶绿素含量及fad2、fad3基因的表达量变化情况进行分析。研究发现:叶绿素含量随着氮、硼施用量的增加呈先升后降,随种植密度增加而降低;fad2、fad3的表达量变化与叶绿素含量规律相同,且通过线性拟合分析发现,fad2、fad3基因表达量与叶绿素含量呈正相关关系。结果表明,fad2、fad3基因在A2B1C2与A2B1C3处理下均有较高表达量,而在A1B3C1、A3B3C1及A1B3C3处理下均有较低表达量,而在较低表达量下,可能有助于其他类脂肪酸(如油酸等)的积累,从而对提高油菜籽品质具有重要意义,有待进一步研究。
The fatty acid composition is one of the important goals to improve rapeseed quality,different environment and cultivation conditions can altered fatty acid composition. This experiment was to promote the improvement of rapeseed fatty acid composition from molecular aspects. Brassica napus 420 was used as material,the design method of ternary quadratic regression orthogonal combination was adopted,chlorophyll content and fad2 and fad3 gene were studied in different periods and different cultivation conditions. And found that: the content of chlorophyll increased first and then decreased with the increased of nitrogen and boron content,but decreased with the increased of density; the changes of the expressions of fad2 and fad3 were similar to those of chlorophyll,besides,it found that there was positive correlation between the amount of chlorophyll content and fad2 and fad3 gene expression by linear regression analysis. In addition,we found that the expression of fad2 and fad3 was higher in A2 B1 C2 and A2 B1 C3,but had lower expression in A1 B3 C1,A3 B3 C1 and A1 B3 C3,that may contribute to the accumulation of other fatty acids( such as oleic acid,etc.) at lower expression,so as to improve the quality of rapeseed is of great significance,it needs to be further studied.
