猪流行性腹泻病毒经典株与变异株高分辨率熔解曲线方法的建立

摘要 为了建立鉴别检测猪流行性腹泻病毒(PEDV)经典株与变异株的高分辨率溶解曲线方法,基于PEDV经典株与变异株在S基因上序列的差异设计1对PCR引物用于RT-PCR扩增,扩增产物通过高分辨率熔解曲线(HRM)分析,建立鉴别PEDV经典株与变异株的PCR-HRM分析方法。通过此方法检测了10份临床样品,PCR-HRM结果表明,4份为PEDV经典毒株,6份为PEDV变异毒株。该方法特异性好,灵敏度高,PCR后无需开盖分析,避免了污染问题,是一种新型的鉴别检测方法。 In order to build a differential diagnosis of high resolution melting curve method of porcine epi- demic diarrhea virus classic and mutant strains, based on the differences in S gene sequence,a pair of prim- ers were designed for RT-PCR amplification, and then high resolution melting-curve(HRM) was estab- lished for differentiating between mutant strains and classic strains. 10 clinical samples were screened by this assay,and 4 positive for classical strains and 6 positive for mutant strains were found.The experiments revealed that the developed RT-PCR and HRM assay was specific and sensitive, and it could be used for rapid and accurate differentiation of PEDV mutant strains and classic strains.
