摘要 目的:探讨基质金属蛋白酶-10(MMP-10)在非小细胞肺癌(NSCLC)组织中的表达、临床意义及其对肺癌A549细胞增殖和转移的影响。方法:采用western blotting法检测60例NSCLC患者肺癌组织及其癌旁组织中MMP-10的表达。设计并合成针对MMP-10基因的特异性小RNA干扰序列(siRNA),转染A549细胞,western blotting法和荧光定量PCR(qPCR)法检测干扰效果。MTT法检测干扰MMP-10的表达对A549细胞增殖的影响,Transwell实验检测干扰MMP-10的表达对A549细胞迁移的影响。结果:MMP-10蛋白在肺癌组织中的表达明显高于癌旁组织(P<0.05),且与患者淋巴结转移有关(P<0.05),而与性别、年龄、吸烟史无关(P>0.05)。干扰组A549细胞MMP-10基因和蛋白表达水平明显低于空白组(未做任何处理)和阴性对照组(转染阴性对照siRNA)。与空白组和阴性对照组比较,MMP-10干扰组A549细胞的增殖、迁移能力显著降低(P<0.05)。结论:NSCLC组织MMP-10蛋白表达明显上调,其表达与患者淋巴结转移有关;下调MMP-10的表达可抑制肺癌A549细胞的增殖和转移。 Objective: To investigate the expression and clinical significance of matrix metalloproteinase-10 (MMP-10) in non-small cell lung cancer (NSCLC) and its effect on the proliferation and metastasis of lung cancer A549 ceils. Methods: The expression of MMP-10 in NSCLC tissues and peficancerous tissues from 60 NSCLC patients was detected. The specific small RNA interference sequences (siRNA) for MMP-10 gene were synthesized and transfected into the A549 cells. The transcription of MMP-10 gene was detected by fluorescence quantitative PCR (qPCR) and the protein expression of MMP-10 was determined by western blotting. MTT as- say was used to investigate the proliferation of the transfected A549 cells, and the cell migration was evaluated by transwell migration assay. Results: The protein expression of MMP-10 in lung cancer tissues was significant- ly higher than that in the peficancerous tissues (P〈0.05), and it was associated to lymph node metastasis (P〈 0. 05), but not correlated with patients' age and history of smoking (P〉0. 05). MMP-10 mRNA and protein levels were significantly decreased after siRNA-MMP-10 transfection into A549 cells, which showed obvious re- duction in the cell proliferation and migration (P〈 0.05). Conclusion: The MMP-10 protein level in NSCLC tissues was significantly up-regulated, and was related to lymph node metastasis. Down-regulation of MMP-10 expression in lung cancer A549 cells could result in lowered cell growth rate and migration ability.
