摘要
[目的]研究青赤散灌肠对溃疡性结肠炎(ulcerative colitis,UC)大鼠血清内毒素(LPS)、肿瘤坏死因子-α(TNF-α)及结肠上皮细胞凋亡的影响,探讨青赤散灌肠治疗UC的可能作用机制。[方法]将40只大鼠随机分为正常组、模型组、中药组及西药组,每组10只。除正常组外,其他3组采用三硝基苯磺酸(TNBS)/乙醇诱导大鼠UC模型。造模后,正常组和模型组大鼠均给予0.9%NaCl注射液5ml/kg灌肠,中药组给予青赤散按生药量19g/kg灌肠,西药组给予美沙拉秦灌肠液(莎尔福)按生药量0.4g/kg灌肠,连续灌肠14d。检测各组大鼠血清LPS、TNF-α含量及结肠上皮细胞凋亡情况。[结果]与正常组比较,给药后模型组大鼠血清LPS、TNF-α含量升高,结肠上皮细胞凋亡增加(P<0.05);与模型组比较,中药组和西药组大鼠血清LPS、TNF-α含量降低,结肠上皮细胞凋亡减少(P<0.05),中药组与西药组疗效相当。[结论]青赤散灌肠可能通过降低UC大鼠血清LPS、TNF-α含量,减少结肠上皮细胞凋亡而起到治疗UC的作用。
[Objective]To observe the effect of Green and red powder enema on serum LPS,TNF-α and apoptosis of colon epithelial cells in UC rats and study the possible mechanism of green and red powder enema in the treatment of UC.[Methods]Totally 40 rats were randomly divided into the normal control group,the model group,the Chinese medicine group and the western medicine group,10 rats in each group.The model of UC rats was prepared by three nitrobenzene sulfonic acid（TNBS）/ethanol.After modeling,0.9% NaCl（5 ml/kg）was administered to rats in the model group and the normal group by enema.Rats in the Chinese medicine group were administered with 19 g/kg crude Green and red powder by enema.Rats in the western medicine group were administered with 0.4 g/kg crude Mesalazine Enemas by enema.The medication lasted for 14 successive days.The contents of serum LPS,TNF-α and the number of apoptotic cells in colonic epithelium in each group were detected.[Results]Compared with the normal group,the content of serum LPS,TNF-α and the number of apoptotic cells in colonic epithelium increasedin the model group after medication（P0.05）.Compared with the model group,the content of serum LPS,TNF-α and the number of apoptotic cells in colonic epithelium were decreased in the Chinese medicine group and the Western medicine group（P0.05）.The curative effect of Chinese medicine group and Western medicine group is equivalent.[Conclusion]Green and red powder enema was a better treatment for US rats possibly by reducing the content of serum LPS,TNF-α and the number of apoptotic cells in colonic epithelium.
