摘要
在模拟人体生理条件下,运用荧光光谱法、同步荧光法和紫外光谱法研究了注射液头孢唑啉钠(CS)与人血清白蛋白(HSA)的相互作用.结果表明:头孢唑啉钠对HSA的内源荧光产生猝灭作用,由Stern-Volmer方程和紫外光谱确定其猝灭过程为静态猝灭,获得了不同温度下的结合常数、结合位点数和热力学参数,并由此推断二者之间通过范德华力和氢键共同作用形成了1∶1的配合物;同步荧光表明头孢唑啉钠对HSA的构象有影响;希尔系数n_H>1,表明CS与HSA作用后存在药物正协同作用.共存物质Hg^(2+)、Pb^(2+)和维生素C对CS-HSA体系的结合常数和结合位点数均有影响.
The interaction between cefazolin sodium(CS)and human serum albumin(HSA)is studied by fluorescence spectroscopy and ultraviolet absorption spectrometry under simulated physiological conditions.The fluorescence spectroscopic analysis of the emission quenching indicates that the quenching mechanism of human serum albumin by cefazolin sodium is a static quenching process,which is in good agreement with the result from ultraviolet absorption spectroscopy.The binding constants,binding sites and thermodynamic paraments are obtained at different temperatures,and it is deduced that the 1∶1 complex is formed by Van der Waals force and hydrogen bond.Synchronous fluorescence indicates that cefazolin sodium has an effect on the conformation of HSA.Hill coefficients(nH〉1)indicates that there exists a certain positive cooperativity effect when CS bound to HSA.Furthermore,the coexisting substances Hg^2+,Pb^2+ and vitamin C has an effect on the interaction between CS and HSA.
