当归四逆汤对ASO寒凝血瘀型家兔下丘脑区功能及相关神经递质的影响

摘要 目的:观察当归四逆汤对ASO寒凝血瘀型家兔下丘脑区功能及相关神经递质的影响。方法:在高脂高胆固醇饲料喂养的基础上,用冰水冷冻法建立寒凝血瘀型模型,采用定位电流刺激股髂动脉法制备动脉硬化闭塞症模型。采用BLOD-f MRI技术对家兔脑功能进行检测,采用酶联免疫法检测脑组织中5-HT、CGRP、YNPY的含量。结果:脑激活区集中在家兔下丘脑部位及附近,正常组、当归四逆汤中剂量组和当归四逆汤低剂量组均有激活,模型组和当归四逆汤高剂量组无激活,正常组与模型组有差异,低剂量、中剂量与高剂量有差异。模型组较正常组下丘脑中5-HT、NPY水平明显升高,CGRP明显降低;与模型组比,当归四逆汤中、高剂量组下丘脑5-HT和NPY的水平明显下降,当归四逆汤低剂量组5-HT明显下降,NPY无明显差异,三个给药组CGRP的水平显著升高。结论:当归四逆汤通过调节ASO家兔下丘脑功能及其相关神经递质水平防治ASO。 Objective: To observe the effect of Danggui Sini Decoction on the function of hypothalamic area and related neurotransmitters in ASO of cold blood coagulation and blood stasis type of rabbits. Methods: Model rabbits were given high fat-high cholesterol feed and frozen in ice water. The model rabbits' femoral iliac arteries were stimulated in current. The brain function of rabbits was detected by BLOD-f MRI technique,and the contents of 5-HT,CGRP and YNPY in brain tissue were detected by enzyme immunoassay. Results: Brain activation areas were concentrated in the hypothalamus of the rabbits. Normal group,middle and low dose groups of Danggui Sini Decoction were activated. The model group and high dose group of Danggui Sini Decoction had no activation. The normal group had differences compared with the model group. Low and middle dose groups had difference compared with high dose group. The model group's 5-HT and NPY were obviously increased than those of the normal group and CGRP obviously decreased. Compared with the model group,middle and high dose groups' 5-HT and NPY were obviously decreased and the low dose group' s 5-HT decreased obviously and NPY had no difference. The three dose groups' CGRP levels were increased obviuosly. Conclusion: The prevention and treatment of ASO by Danggui Sini Decoction is regulating the function of hypothalamus and the related neurotransmitters in the hypothalamus of the ASO.
