响应面优化海带岩藻黄素超声提取工艺研究 被引量:4

Optimization of Ultrasound-Assisted Extraction Technology of Fucoxanthin from Kelp by Response Surface Methodology

摘要 研究主要为探究超声辅助提取海带岩藻黄素的最佳工艺参数。试验以新鲜海带为原料,优化确定最佳提取剂及原料干制方式。通过单因素试验与响应面试验,以岩藻黄素得率为响应值,优化超声提取工艺。试验确定最佳提取试剂为80%乙醇,最佳干制方式为40℃烘干24 h。最优提取工艺参数为:提取时间66.8 min,提取温度60.9℃,液料比值24.9 m L/g。该工艺操作简单,稳定可靠,可重复性强,且安全无污染。岩藻黄素得率达0.610 1 mg·g^(-1),与响应面预测值0.599 7 mg·g^(-1)基本一致。 The purpose of this study was to optimize ultrasound-assisted extraction technology of fucoxanthin from kelp. Extracting agent and drying type were optimized. On the basis of single factor test, the response surface methodology was used to investigate the effects of process in processing parameters, including extraction time, temperature and solid-liquid ratio. The extraction rate of fucoxanthin was chosen as response value. Results showed that optimum extracting agent was 80% alcohol and 40℃ drying for 24 h was optimum. The optimum technological condition was that extraction time of 66.8 min, extraction temperature of 60.9 ℃, liquid-solid ratio of 24.9 mL/g. Under the above condition, extraction ratio of fucoxanthin was up to 0.610 1 mg·g-1, which was highly consistent with the predictive value of 0.599 7 mg·g-1.
