饿蚂蝗总黄酮对鸭乙型肝炎的保护作用研究 被引量:4

摘要 目的:研究饿蚂蝗总黄酮对鸭乙型病毒性肝炎的作用。方法:采用鸭乙型肝炎病毒(DHBV)血清感染1日龄广西麻鸭。感染7d后采用PCR法筛选出DHBV DNA强阳性鸭,随机分为5组:模型组、阳性药拉米夫定组(50mg/kg)、饿蚂蝗总黄酮组(300,150,75 mg/kg),每组10只。另设正常对照组(经PCR筛选出的未造模DHBV阴性鸭)10只。各组从造模后第14d开始给药,每天灌胃给药1次,持续给药14d。每组动物分别于给药前0天(T0)、给药后7天(T7)、给药后14天(T14)、停药后3天(P3)自颈静脉采血,检测血清中鸭乙型肝炎病毒DNA(DHBV DNA)、鸭乙型肝炎表面抗原(DHBs Ag)和鸭乙型肝炎e抗原(DHBe Ag)的水平,谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性,白介素-2(IL-2)、γ干扰素(IFN-γ)的含量变化。结果:与模型组比较,在T14时,饿蚂蝗总黄酮300、150、75 mg/kg剂量组能够降低T14时鸭血清DHBV DNA载量以及ALT和AST含量,升高IL-2和IFN-γ水平;在P3时,饿蚂蝗总黄酮300、150mg/kg剂量组对鸭血清DHBV DNA载量以及ALT和AST含量的抑制仍然明显,停药后无反跳。结论:饿蚂蝗总黄酮具有抗鸭乙型病毒性肝炎作用,其机制与抑制病毒复制、保肝降酶、调节免疫功能有关。 Objective: To study the effect of total flavonoids from Desmodium multiflorum DC. on DHBV hepatitis. Methods: One-day-old ducklings were infected with duck hepatitis B virus( DHBV). Strong positive ducklings DHBV were detected by PCR assay after 7 days' infection,and were randomly divided into 5 groups,with each consisting of 10 ducklings: the model group,50 mg/kg lamivudine group,total flavonoids from Desmodium multiflorum DC. groups( 300,150,75 mg/kg). 10 DHBV negative ducklings were served as the control group. Drugs were orally given daily for 14 days. The blood was drawn from jugular vein of all ducks before the treatment( T0),after the medication for 7 days( T7),14 days( T14) and 3 days after withdrawal of the drug( P3). Levels of DHBV DNA,DHBs Ag,DHBe Ag,ALT,AST,IL-2 and IFN-γ were detected. Results: Compared with the model group,total flavones in Desmodium multiflorum groups( 300,150,75 mg/kg) significantly decreased levels of DHBV DNA,ALT and AST,significantly increased contents of IL-2 and IFN-γ at T14. Total flavonoids from Desmodium multiflorum DC. significantly decreased levels of DHBV DNA,ALT and AST,and didn't observe the rebound after 3 days of suspension. Conclusion: Total flavonoids from Desmodium multiflorum DC. has the therapeutics effect on DHBV hepatitis,and its mechanism may be contributed to antiviral,hepatic-protective,descending transaminase function and immune adjustment.
