摘要
目的:研究牛黄降压胶囊的降压活性、降压特点,安全性及长期给药对自发性高血压大鼠(SHR)心肝肾的影响,并初步揭示降压作用机制。方法:32只雄性SHR随机分为模型对照组,牛黄降压胶囊组,牛黄降压丸组,牛黄降压片组;另设WKY鼠作为正常对照组。各组连续灌胃给药31天,牛黄降压胶囊、牛黄降压丸及牛黄降压片组给药剂量分别为0.32 g/kg,0.64g/kg和0.30g/kg体重,于不同时间点测定血压。治疗结束后,取正常对照组、模型组和牛黄降压胶囊组大鼠心脏、肾脏、脾脏及肝脏,进行病理学分析。测定各组动物肝肾功能ALT、AST、BUN和CREA指标;测定肾脏及血浆中肾素、血管紧张素II含量。结果:0.32 g/kg牛黄降压胶囊和0.30g/kg牛黄降压片在给药第6天显示出明显降压效果;三种剂型均在给药21天后达到最大降压效果,且于一天内不同时间点降压效果保持稳定;治疗结束48h后牛黄降压胶囊仍保持显著降压效果。治疗期间,牛黄降压胶囊对于肝肾功能未见显著性影响。病理学结果显示,牛黄降压胶囊对高血压引起的心肌肥厚和肾小球毛细血管的高灌注具有一定的防治作用。牛黄降压胶囊可显著降低高血压动物血浆中的肾素和血管紧张素II水平。结论:牛黄降压胶囊降压疗效确切,降压平稳、长效,对高血压引起的心血管并发症有一定的防治作用。
Objective： To evaluate the effect,characteristics and the safety in spontaneously hypertensive rats（ SHR） of Niuhuang Jiangya（ NHJY）capsule,a traditional Chinese medicine used in china to treat hypertension,then to evaluate protective effect on the secondary injury of heart and kidney in hypertension,and to initially reveal the antihypertensive mechanism. Methods： Thirty-two SHR rats（ male） were randomly divided into four groups： the model group,NHJY capsule group,NHJY pill group and NHJY tablet group,while another 8 WKY rats were set as the normal group. All rats were treated with NHJY through intragastric administration for 31 days,and doses of NHJY capsule,NHJY pill and NHJY tablet were 0. 32 g/kg,0. 64 g/kg and 0. 30 g/kg body weight,respectively. The blood pressure was measured at different time points,and at the end of treatment,formalin-fixed tissues from heart,kidney,liver and spleen in the WKY,model and NHJY capsule were embedded in paraffin,and stained with hematoxylin-eosin（ HE）. Levels of ALT,AST,BUN,CREA,rennin and angiotensin II were measured,respectively. Results： 0. 32 g/kg NHJY capsule and 0. 30 g/kg NHJY tablet showed the obviously antihypertensive effect on the 6 th day of administration. Three preparation forms reached the maximum effect after 21 days＇ administration,and the effect remained during the whole day,48 h after the treatment NHJY capsule still preserved antihypertensive activity. During the treatment,the NHJY capsule had no significant influence on liver and kidney functions. Pathological results showed that the NHJY capsule on cardiac hypertrophy and glomerular expansion induced by hypertension has a positive effect. NHJY capsule significantly reduced levels of renin and angiotensin II in plasma of hypertensive model animals. Conclusion： The NHJY capsule can reliably and steadily reduce the blood pressure of SHR rats,while have a certain protective effect on the cardiovascular complications caused by hypertension.
