摘要
目的探讨精草润目液对干眼大鼠模型角膜上皮和泪液分泌量的影响,为精草润目液的临床使用提供实验依据。方法清洁级SD雌雄各半大鼠60只随机分为两组,分别制作角膜上皮损伤干眼模型和泪液分泌低下干眼模型,造模成功后将各模型大鼠再随机分为空白对照组(予自制滴眼液溶剂)、阳性药物对照组(角膜上皮损伤干眼模型予0.5%羧甲基纤维素钠滴眼液,泪液分泌低下干眼模型予0.2%卡波姆滴眼液)以及精草润目液低、中、高剂量组。角膜上皮损伤模型大鼠以角膜上皮损伤评分为观察指标,泪液分泌低下模型大鼠以泪液分泌量为观察指标,均于治疗7、14、21 d时比较不同干预组的指标差异。结果在角膜上皮损伤干眼模型大鼠,治疗21 d后,高剂量组的角膜损伤评分明显低于阳性药物治疗组(P<0.05)。在泪液分泌低下干眼模型大鼠,治疗21 d后,高、中剂量组泪液分泌量明显高于阳性药物对照组(P<0.05),而低剂量组的泪液分泌量各时间点始终低于阳性药物对照组(P<0.05)。结论精草润目液能明显增加大鼠干眼模型的泪液分泌量,并减轻角膜上皮损伤,药物疗效与药物的浓度及治疗时间相关。
OBJECTIVE To explore the effect of Jingcao eye drops oncorneal epithelium and tear secretion of dry eye model and consequently to provide experimental basis for the clinical use of Jingcao eye drops. METHODS The 60 clean SD rats of half male and half female ones were randomly divided into dry eye due to epithelial injury group and dry eye due to insufficient tear group. After successful modeling, both groups were randomly divided into solvent control group, positive drug control group as well as low dose, medium and high dose of Jingcao eye drops groups. Seven, twelve, twenty-one days after the intervention, result of corneal epithelial damage score（CEDS） and Schirmer test of two groups were evaluated. RESULTS For dry eye due to epithelial injury groups, 21 days after treatment, CEDS of high dose of Jingcao eye drops group was significantly different compared with positive drug con-trol group（P〈0.05）. For dry eye due to insufficient tear groups, result of Schirmer test in high and medium dose of Jingcao eye drop group were higher than positive control group 21 days after treatment while result of low dose group were all lower than positive control group at three time points, and the differences were of statistical significance（P〈0.05）.CONCLUSIONS Jingcao eye drops effectively improved the tear secretion, reduced corneal epithelial damage and its curative effect was associated with concentration and the treatment duration.
