The Effect of EGCG on Myocardial Cell Apoptosis in Mice induced by DOX

摘要 目的:探讨表没食子儿茶素没食子酸酯(Epigallocatechin-3-gallate,EGCG)对阿霉素(DOX)注射引起的小鼠心肌细胞凋亡的影响及其作用机制研究。方法:应用DOX腹腔注射作用于小鼠,建立小鼠体内心肌损伤模型,观察小鼠生理状况,比色法测定细胞乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)的活性及丙二醛(MDA)的含量,利用流式细胞仪检测细胞线粒体膜电位及细胞凋亡状况。结果:与对照组相比,DOX组中LDH和CK的活性显著增强;与DOX组相比,EGCG+DOX组中LDH和CK的活性显著减弱,抑制DOX心肌细胞凋亡。同时EGCG显著减轻DOX引起MDA含量的增加,提高细胞中SOD的活性。线粒体膜电位数据显示,与DOX组相比,EGCG显著增加心肌细胞的线粒体膜电位。结论:EGCG对DOX引起的心血管损伤具有显著的保护作用,其保护作用与降低心肌细胞凋亡,提高抗氧化防御和保护线粒体有关。 Objective: To investigate protective effect of EGCG(epigallocatechin-3-gallate) on myocardial damage induced by injection of doxorubicin(DOX) in mice and mechanisms of mitochondria.Methods: We applied DOX by intraperitoneal injection to set up myocardial damage model and observed physiological status in mice.Then the activities of LDH,CK,SOD and the level of MDA by colorimetric method were measured.And mitochondrial membrane potential and the status of cell apoptosis by flow cytometry were detected.Results: Compared with Control group,the activities of LDH and CK were significantly increased in DOX group.And compared with DOX group,the activities of LDH and CK were significantly decreased and DOX-induced myocardial cell apoptosis was inhibited.Meanwhile,EGCG significantly decreased the level of MDA caused by DOX and improved the activity of SOD in cells.The datum of mitochondrial merabrane potential showed that EGCG significantly increased mitochondrial merabrane potential of myocardial cell in comparion with DOX group.Conclusions: EGCG significantly has protective effect on cardiovascular injuries caused by treatment of DOX.And the protective effect is associated with lowering mycardial cell apoptosis,improving antioxidative defense and protecting mitochondria.
