摘要
建立不同产地蝉花Cordyceps cicadae的高效液相指纹图谱,为蝉花药材的质量控制提供参考。采用HPLC-PAD检测法建立不同产地9批蝉花药材的指纹图谱,采用Agilent Eclipse XDB-C_(18)柱(4.6 mm×250 mm,5μm),流动相乙腈-水梯度洗脱,柱温25℃,检测波长260 nm,体积流量0.5~0.8 m L/min;采用SAS8.0对9个批次蝉花进行聚类分析。对9个批次的蝉花药材进行测定,采用中药指纹图谱相似度评价系统(2004A版)软件分析,以虫草素、腺苷、肌苷、麦角甾醇为参照峰,从中标定了14个共有峰,并根据共有峰对9批次蝉花进行相似度分析和聚类分析。相似度结果表明,9批蝉花样品指纹图谱与共有模式之间相似性良好,除S5与S8之外,其他7个批次蝉花相似度均大于0.95;聚类分析将9批次样品分为两类,S8为第一类,其他均为第二类,相似性结果与系统聚类分析结果基本一致。该方法简便可靠,可以作为蝉花药材质量的评价方法。
To establish an HPLC-fingerprint method for evaluating the quality of the root of Cordyceps cicadae from different habitats in China,the HPLC analysis was performed on an Agilent Eclipse XDB-C_(18)(4.6 mm × 250 mm,5 μm) with an acetonitrile-water solution in gradient elutionas mobile phase.The flow rate was 0.5 ~ 0.8 mL/min.The detective wavelength is 260 nm.The column temperature is 25 ℃.Then,the data from HPLC fingerprint were analyzed by similarity evaluation,principle component analysis,and system cluster analysis.The similarities of the 7 batches of samples were above 0.95 except for S5 and S8.Cluster analysis divided the 9 samples into two batches,S8 was the first,and the others were the second.The results of the similarity analysis and the clustering analysis are basically the same.The method is simple,reliable,and it can provide a standard and guidance for the quality control of Cordycepscicadae.
