基于Fe_3O_4纳米材料的还原型谷胱甘肽传感器对丙烯酰胺的检测 被引量:1

摘要 丙烯酰胺(AA)是一种能够对人体产生神经毒性、生殖发育毒性、遗传毒性以及致癌性的有害物质,并且高淀粉、高蛋白食品经高温处理后均能产生,所以建立一种快速检测AA的方法至关重要。人体中AA的代谢途径主要是与还原型谷胱甘肽(GSH)或血红蛋白(Hb)发生加成反应。GSH在空气中容易发生氧化还原反应,因此对AA的电化学检测大多采用Hb修饰的生物传感器。Fe_3O_4纳米粒子具有较好的生物相容性,能够与GSH中的—COOH基团结合,不影响GSH的氧化性能,并且能够提升GSH的电信号。制备了Nafion/GSH-Fe_3O_4/GC传感器,采用循环伏安法对实际样品中的AA进行定性定量检测。通过对试验条件的优化,确定最佳条件为p H 4.5、GSH修饰量100μg、加成反应时间60 s。在最佳条件下,对不同浓度的AA进行检测,建立标准曲线,确定线性范围为1×10-7~1×10-5 mol/L,线性方程为Y=-0.299X+2.699(R2=0.996 7),样品回收率为100.45%。该方法实现了对丙烯酰胺的快速检测,并且对GSH应用于检测AA提供了理论基础。 Acrylamide(AA) is a kind of harmful substance which can produce neurotoxicity,reproductive development toxicity,genetic toxicity and carcinogenicity to human body. AA can be produced in high starch and protein food after high temperature treatment,so it is very important to establish a method for rapid detection of AA. The main metabolic way of AA in human body is that acrylamide(AA) reacted with reduced glutathione(GSH)or hemoglobin(Hb). GSH is easy to undergo redox-reaction in air,so Hb-modified biosensor is mostly used in the electrochemical detection of AA. Fe_3O_4 nanoparticles have good biocompatibility and can be combined with the —COOH group in GSH,which does not affect the oxidation performance of GSH,and can improve the electrical signal of GSH. Nafion/GSH-Fe_3O_4/GC sensors were prepared and the AA in real samples was detected qualitatively and quantitatively by cyclic voltammetry in the present study. The optimum detection conditions of AA were as follows:p H of 4.5,the amount of modified GSH of 100 μg and reaction time of 60 s.Under the optimal conditions,the AA of different concentrations was detected to establish the standard curve. The linear response w as in the range of 1 ×10-7~1 ×10-5,the linear regression equation was Y=-0.299 X+2.699(R2=0.9967),and the recovery rate of the sample was 100.45%. The rapid detection of acrylamide has been realized by Nafion/GSH-Fe_3O_4/GC sensors method,which will provide a theoretical basis for the electrochemical detection of AA.
