摘要
目的探究艾灸天枢、上巨虚治疗腹泻型肠易激综合征(diarrhea predominant irritable bowel syndrome,D-IBS)的机制。方法将30只SD大鼠随机分为空白对照组、模型组、艾灸组,每组10只。采用束缚加番泻叶煎剂灌胃的方法复制D-IBS模型。艾灸组大鼠每日艾灸双侧天枢、上巨虚20min,连续7d。采用ELISA法检测大鼠血清白细胞介素-6(interleukin-6,IL-6)水平,分别采用Western blot法、RT-PCR法检测大鼠结肠组织β-防御素-2(beta-defensin-2,BD-2)蛋白及其mRNA的相对表达水平。结果艾灸能够显著降低D-IBS大鼠升高的血清IL-6水平和结肠组织BD-2蛋白及其mRNA表达水平(P<0.05)。结论艾灸天枢、上巨虚治疗D-IBS的机制可能与TLR4/NF-κB信号通路有关。
Objective To investigate the mechanism of moxibustion at Tianshu and Shangjuxu in the treatment of diarrhea-predominant irritable bowel syndrome(D-IBS).Methods A total of 30 Sprague-Dawley rats were randomly divided into blank control group,model group,and moxibustion group,with 10 rats in each group.A rat model of D-IBS was established by restraint and senna decoction given by gavage.The rats in the moxibustion group were given moxibustion at Tianshu and Shangjuxu on both sides 20 minutes a day for7 consecutive days.ELISA was used to measure the serum level of interleukin-6(IL-6),and Western blot and RT-PCR were used to measure the relative protein and mRNA expression of β-defensin-2(BD-2)in the colon tissue.Results Moxibustion significantly reduced the increased serum level of IL-6 and the increased protein and mRNA expression of BD-2 in the colon tissue in D-IBS rats(P<0.05).Conclusion The TLR4/NF-κB signaling pathway may involved in the mechanism of moxibustion at Tianshu and Shangjuxu in the treatment of D-IBS.
