摘要
目的检测Tim-3在早期自然流产患者外周血中的表达及其与Th17/Treg的关系,探讨其在早期自然流产中的作用。方法选取正常早孕自愿人工流产患者36例为观察组,早期自然流产患者41例为研究组,利用流式细胞检测技术检测2组患者外周血中Tim-3、Th17、Treg的表达,其中流式细胞仪检测CD4细胞中IL-17(CD4+IL-17+)比例反映Th17水平,CD4细胞中CD4+CD25+Treg反映Treg细胞水平,并探讨Tim-3的表达及与Th17/Treg的关系。结果 Tim-3、Th17及Th17/Treg在研究组升高,观察组降低;Treg在研究组降低,观察组升高,2组比较有显著性差异。结论 Tim-3的异常表达可能预示着自然流产的发生,与Th17/Treg失衡有关。
Objective：To detect the expression of T cell immunoglobulin mucin 3（Tim-3）and its relationship with Th17/Treg cytokine from peripheral blood mononuclear cells（PBMCs）,to investigate the role of Tim-3 in the occurrence in the the early spontaneous abortion（ESA）.Methods：41 pregnant women suffered with ESA were recruited as the study group and 36 normal pregnant women were chosen as the control group.For the two groups,Tim-3,Th17 and Treg in PBMC were detected by flow cytometry,Levels of CD4＋IL-17＋ and CD4＋CD25＋Treg,reflecting expressions of Th17 and Treg respectively.Results：Contrast with the normal control group,the expression of Tim-3、Th17 and Th17/Treg ratio in PBMCs in the study group increased significantly（P〈0.05）;the expression of Treg in PBMCs compared with those in the normal control group decreased（P〈0.05）.Tim-3 expression in PBMCs was changed in consistency with Th17/Treg ratio.Conclusions：Tim-3 level in PBMCs in study group was increased,indicating that Tim-3 might take part in the occurrence of the early spontaneous abortion,has a very close relationship with the Th17/Treg imbalance.
