乌药醇提取物抗慢性酒精性肝损伤量效关系研究

摘要 目的观察不同剂量乌药醇提取物对慢性酒精性肝损伤进展中血生化指标及炎症因子水平的影响,探讨乌药醇提取物抗慢性酒精性肝损伤的量效关系。方法乙醇灌胃复制慢性酒精性肝损伤小鼠模型,予以不同剂量的乌药醇提取物干预;在第2、4、6N采血,用生化仪检测血清肝功能指标、血脂水平;在第4、6周,用ELISA法检测血清IL-6、IL-1β水平。结果2周后,模型小鼠血清ALT、AST活性随着造模时间的延长而显著增加,血清TC、HDL-C水平随着造模时间的延长而逐渐降低;第4、6N血清IL-6、IL-1β水平均显著上升。给药4周后,2g/kg乌药组能降低ALT活性,2g/kg、1g/kg、0.5g/kg乌药组能降低AST活性;6周后1g/kg、0.5g/kg、0.2g/kg乌药组对ALT活性降低明显,1g/kg、0.5g/kg、0.2g/kg、0.1g/kg乌药组能显著降低血清AST活性。给药组对血脂影响较小。大多数浓度乌药醇提物组对IL-6、IL-18水平有降低作用。结论乌药醇提取物具有抗慢性酒精性肝损伤作用,但不同剂量对抗慢性肝损伤的效果、侧重点有差异。 Objective To observe the changes of biochemical indexes and inflammatory factor level in progression of chronic alcoholic liver injury (ALI ) and different doses of ethanol Radix Linderae extraction ( RLEE ) against chronic liver injury. Methods All models were established by gavage ICP. mice with gradient alcoholic solution, and at the same time, mice in the experiment groups were orally administered with different doses of Linderae Radix by alcohol extract for six weeks. At 2, 4, 6 week, the blood of mice was collected, serum ALT, AST, TC, TG, HDL-C were determined by automatic biochemical analyser and serum IL-6, IL-1 β were determined by ELISA according to the specification of the kits. Results After 2 weeks, the activities of ALT and AST in all models were increased according to the prolong of the ethanol intake.With the increase of molding time, the serum TC, HDL-C levels gradually decreased. Levels of serum IL-6, IL-1β were observed at 4w and 6w.At 4w RLEE about 2g/kg could lower serum ALT activities, RLEE about 2g/kg, lg/kg, 0.Sg/kg could lower serum AST activities.At 6w the doses of lg/kg, 0.Sg/kg, 0.2g/kg RLEE could lower serum ALT significantly.RLEE about 1 g/kg, 0.Sg/kg, 0.2g/kg, 0.1 g/kg doses could significantly lower the activity of AST. Each dose of RLEE had no obviously changed about serum hpid levels. Most of doses of P.LEE could decrease the levels of serum IL-6 and IL-1 levels. Conclusion The ethanol Radix Linderae extraction has the effect of anti chronic hver injury, but different doses against chronic liver injury has different effects.
