摘要
目的:探讨重性抑郁障碍(MDD)患者外周血单个核细胞(PBMC)中微小RNA(miR)-132表达水平在抗抑郁治疗前后的改变。方法:给予32例MDD患者(MDD组)8周的抗抑郁药治疗;以治疗后汉密尔顿抑郁量表24项(HAMD-24)评分的减分率≥50%为有效;治疗前后采用实时定量反转录-聚合酶链反应(qRT-PCR)对MDD组患者PBMC中miR-132表达进行定量分析;并与30名健康对照者(对照组)比较。结果:基线期MDD组PBMC中miR-132相对表达量与对照组比较差异无统计学意义(P=0.226);治疗后MDD组PBMC中miR-132相对表达量较基线明显上调(t=-2.356,P=0.025);且有效患者miR-132表达水平较基线明显升高(t=-2.804,P=0.01),无效患者治疗前后PBMC中miR-132表达水平比较差异无统计学意义(z=-0.943,P=0.438)。结论:MDD患者PBMC中miR-132表达水平的变化可能与抗抑郁药的疗效有关。
Objective: To investigate the expression changes of microRNA-132( miR-132) in the peripheral blood mononuclear cells( PBMCs) in patients with major depressive disorder( MDD) before and after treatment. Method: Thirty-two MDD patients received antidepressant for 8 weeks. After treatment,the ratio of score reduce of 24-item Hamilton depression rating scale( HAMD-24) over 50% was defined as effective.Real-time quantitative reverse transcription-polymerase chain reaction( qRT-PCR) was applied to quantitative analysis the expression of PBMC miR-132 before and after treatment in MDD patients; and the results were compared with 30 healthy controls( control group). Results: There was no significant difference of the relative expression of PBMCs miR-132 between baseline of the MDD group and the control group( P = 0. 226). After treatment,the relative expression of PBMCs miR-132 was significantly up-regulated in MDD group( t =-2. 356,P = 0. 025). Furthermore,the level of PBMCs miR-132 was increased significantly in responder patients( t =-2. 804,P = 0. 01),but was not increased significantly in non-responder patients( z =-0. 943,P =0. 438). Conclusion: The change of PBMC miR-132 expression in MDD patients may be associated with antidepressant effect.
