体外人脐带间充质干细胞成软骨定向诱导分化的实验研究

摘要 目的探讨体外人脐带间充质干细胞(MSCs)向软骨细胞分化的可能性。方法取足月剖官产健康新生儿脐血,应用贴壁法分离获取MSCs,观察其形态学特点并检测细胞表面标志,体外定向成软骨分化诱导培养3周。结果人脐带MSCs表达CD44,不表达CD45,经成软骨诱导分化3周后呈细胞呈多角形改变,番红O染色、Ⅱ型胶原染色检测均为强阳性,提示分泌胞外基质糖胺多糖和Ⅱ型胶原。结论体外人脐带MSCs可以定向诱导分化成软骨细胞,有望成为关节软骨组织工程的新型种子细胞。 Objective To investigate the possibility of differentiation of human umbilical cord mesenchymal stem cells(MSCs) into chondrocytes in vitro. Methods Human umbilical cord blood was obtained from healthy newborns with full term cesarean section, and MSCs was isolated by adherence method. Morphological characteristics and cell surface markers were observed, and chondrogenic differentiation was induced in vitro for 3 weeks. Results MSCs were stained positive for MSCs marker CD44 but negative for hematopoietic CD45. After 3 weeks of induction, oil red-O staining and type Ⅱ collagen staining were positive, suggesting that the secretion of extracellular matrix glycosaminoglycans and collagen type Ⅱ. Conclusion Human umbilical cord MSCs can be induced to differentiate into chondrocytes in vitro, which is expected to become a new type of seed cell for articular cartilage tissue engineering.
