BAK/cFLIP在女性压力性尿失禁患者尿道周围肌细胞中的表达及意义

Expression and significance of BAK and cFLIP in urethral peripheral muscle cells of women with stress urinary incontinence

摘要 目的探讨凋亡相关基因BAK/c FLIP与压力性尿失禁(SUI)的关系。方法随机选择资料完整的20例女性因SUI行手术治疗患者为观察组,其中单纯SUI组15例,SUI+盆腔器官脱垂(POP)组5例;同期选择20例无SUI患者为对照组。采用免疫组化和q RI-PCR方法检测各组阴道前壁组织中BAK、c FLIP基因蛋白的表达情况。结果BAK在SUI组、SUI+POP组、对照组中的表达阳性率分别为66.67%、60.00%、10.00%,在3组中的基因表达量分别为0.0045±0.0042、0.0048±0.0045、0.0011±0.0009;c FLIP在3组中表达阳性率分别为26.67%、20.00%、85.00%,在3组中的基因表达量分别为0.0012±0.0011、0.0010±0.0011、0.0056±0.0048。SUI组和SUI+POP组分别与对照组比较,BAK和c FLIP阳性表达率及表达量均有统计学差异(P<0.05),SUI组与SUI+POP组比较,差异均无统计学意义(P>0.05)。BAK与c FLIP表达水平呈负相关(r=-0.211,P<0.05)。结论 BAK和c FLIP的负相互作用,可能参与了尿道周围肌组织平滑肌细胞凋亡与增殖过程,在压力性尿失禁的诊断和治疗中具有重要意义。 Objective To investigate the relationship between apoptosis related gene BAK/c FLIP and stress urinary incontinence. Methods Twenty female patients with SUI and intact follow-up data were randomly selected as observation group, 15 cases of pure SUI group, 5 cases of SUI+POP group. In the same period,20 patients without SUI were selected as control group. SP method and q RT-PCR were performed to detect the expressions of BAK and c FLIP in vaginal anterior wall tissue. Results The positive rate of BAK in SUI group, SUI+POP group and control group were 66.67%,60.00% and 10.00%.The number of gene expression in the three groups were 0.0045±0.0042,0.0048±0.0045 and 0.0011±0.0009. The positive rate of c FLIP in the three groups were 26.67%, 20.00%,85.00%.The number of gene expression in the three groups was 0.0012±0.0011,0.0010±0.0011 and 0.0056±0.0048. Compared with control group respectively,the expressions of BAK and c FLIP had statistically significant differences in the SUI group and the SUI+POP group(P〈0.05).But there was no significant difference between the SUI group and the SUI+POP group in the expressions of BAK and c FLIP(P〈0.05).The expression of BAK was negatively correlated with c FLIP(r=-0.211, P〈0.05). Conclusion The negative interaction between BAK and c FLIP may be involved in the apoptosis and proliferation of muscle cells around the urethra, which is important in the diagnosis and treatment of stress urinary incontinence.
