摘要
根据Gen Bank数据库中禽白血病病毒(ALV)K亚群特异性的gp85基因序列,设计了特异性检测ALV K亚群(ALV-K)的RT-LAMP引物。对内、外引物浓度组合和反应温度进行优化,建立了ALV-K的RT-LAMP检测方法。特异性试验结果显示,本方法只对ALV-K进行特异性扩增,而对禽白血病的其他亚群以及NDV、REV、AIV、MDV、IBV等常见禽病病原未见扩增。敏感性试验表明,本方法最低能够检测到3.4×104copies/L的质粒。
Based on the gp85 gene sequences of avian leukosis virus subgroup K fromGenBank database, the specific RT-LAMPprimers were designed to detect avian leukosis virus subgroup K (ALV-K). The dif- ferent concentration combinations of inner and outer primers were tested and reaction temperature was optimized to establish the RT-LAMPmethod for detecting ALV-K. The results showed that this method can specifically detect ALV-K without cross-reactions with other subgroups of ALV or other poultry disease pathogens such as NDV,REV,AIV,MDV,IBV,etc..The sensitivity test indicated that the minimum detectable limit was 3.4× 104copies of plasmids.
