钙敏感受体与含钙肾结石形成的相关性 被引量:7

The correlation between CaSR and calcium nephrolithiasis formation

摘要 目的通过检测含钙肾结石(CN)患者肾组织中钙敏感受体(CaSR)、受体活性修饰蛋白(RAMP)、小凹蛋白(caveolin)以及细丝蛋白A(FLNA)的表达,探讨CaSR与CN形成的相关性。方法选取手术切除的CN患者无功能肾组织30例为观察组,另选取同期行根治性肾切除术肾癌患者远癌正常肾组织30例为对照组,通过Real-time PCR、Western blot和免疫组化SABC法检测CaSR、RAMP、caveolin、FLNA mRNA及蛋白表达。结果观察组CaSR mRNA及蛋白表达均低于对照组(P<0.05),而RAMP、caveolin、FLNA mRNA及蛋白表达均高于对照组(P<0.05)。Spearman相关性分析显示,CaSR蛋白表达与caveolin、RAMP、FLNA蛋白表达均呈负相关(P<0.05)。结论CaSR表达降低可能促进CN的形成,且caveolin、RAMP、FLNA可能通过调节CaSR的表达参与了CN的形成。 Objective To investigate the expression of calcium sensing receptor ( CaSR), receptor activity modif- ying protein (RAMP) , caveolin and filamin A (FLNA) in kidney tissue of patients with calcium nephrolithiasis (CN), and to explore the correlation between CaSR and CN. Methods From March 2016 to May 2017, 30 cases of no - func- tion kidneys were collected after the nephrectomy as the case group, and 30 cases of normal renal tissues far away from the tumor were collected as the control group after the radical nephrectomy. CaSR, caveolin, RAMP and FLNA mRNA ex- pression was assessed by Real - time PCR. Expression of CaSR, eaveolin, RAMP and FLNA protein was assessed by Western blot and immunohistoehemistry (SABC). Results In case group, the mRNA expression and the protein expres- sion of caveolin, RAMP and FLNA were significantly higher ( P 〈 0. 05 ) , while CaSR was significantly lower ( P 〈 0. 05 ) , compared with control group. Spearman correlation analysis showed the expression of CaSR protein was significant- ly negatively correlated with caveolin, RAMP and FLNA ( P 〈 0. 05 ). Conclusion The down - regulation of CaSR ex- pression may promote the formation of CN; and caveolin, RAMP and FLNA may be involved in the formation of CN via regulating the expression of CaSR.
