摘要
目的:研究五味消毒饮抑制人皮肤鳞状细胞癌A431细胞增殖及促进凋亡的作用机制。方法:选取人表皮癌A431细胞株作为实验材料,将五味消毒饮作用于体外培养的人皮肤鳞状细胞癌A431细胞,设空白对照组、研究组,于培养细胞后的24 h、48 h、72 h,采用MTT法检测细胞抑制率,观察A431细胞形态,运用免疫组化法检测人皮肤鳞状细胞癌A431细胞的Bcl-2、Bax表达,并用流式细胞仪对细胞凋亡状况进行分析。结果:研究组人皮肤鳞状细胞癌A431细胞培养24h、48 h、72 h后的增殖抑制率均高于空白对照组,且随着时间延长细胞增殖抑制率不断增加,差异有统计学意义(P<0.05);形态学方面,空白对照组人皮肤鳞状细胞癌A431细胞表现为多边形、不规则形,折光性表现强,均匀分布,贴壁生长,细胞生长状态良好;研究组人皮肤鳞状细胞癌细胞A431生长表现不佳,细胞体积缩小,皱缩变圆,周围细胞的黏附力降低,细胞间的间隙扩大,随着培养时间延长,细胞不断脱落漂浮或死亡,细胞数明显减少;研究组Bcl-2细胞阳性率低于空白对照组,Bax细胞阳性率高于空白对照组,差异有统计学意义(P<0.05);流式细胞学检测显示,研究组A431细胞凋亡率高于空白对照组,差异有统计学意义(P<0.05)。结论:五味消毒饮能减少Bcl-2表达,促进Bax表达,抑制人皮肤鳞状细胞癌A431细胞增殖,促进A431细胞凋亡,控制或抑制人皮肤鳞状细胞癌的发生、发展。
Objective: To study the effect of Wuwei Xiaodu drink on proliferation inhibition and promoting apoptosis of skin squamous cancer cell( A431). Methods: The human epidermal carcinoma A431 cell line was selected as the experimental material,and the flavonoid disinfection drink was used to treat the human skin squamous cell carcinoma A431 cells cultured in vitro. The blank control group and the study group were set as control group,being cultured for 24 h,48 h and 72 h respectively. The cell inhibitory rate was measured by MTT assay. The morphology of A431 cells was observed. The expression of Bcl-2 and Bax in human skin squamous cell carcinoma A431 cells was detected by immunohistochemistry. The cell apoptosis was analyzed with flow cytometry.Results: The inhibitory rates of proliferation of A431 cells cultured in human skin squamous cell carcinoma 24 h,48 h and 72 h respectively of the study group were significantly higher than that of the blank control group respectively,and the inhibitory rate of cell proliferation significantly increased with the prolongation of time( P〈0. 05). Morphologically,the A431 cells of the human skin squamous cell carcinoma in the blank control group showed polygons,irregular shape,strong refraction,uniform distribution,adherent growth and good cell growth state. The human skin squamous cell carcinoma cells A431 manifested in poor growth performance,cell volume reduction,shrinkage round,the surrounding cells reduce the adhesion,the gap between cells expanded,with the incubation time,the cells continue to fall off or die and the number of cells was significantly reduced. Flow cytometry showed that the apoptosis rate of A431 cells in the study group was significanlty higher than that in the blank control group( P〈0. 05).The positive rate of Bax cells in the study group was significantly higher than that in the blank control group( P〈0. 05). Conclusion: Wuwei Xiaodu Drink can reduce the expression of Bcl-2,promote the expression of Bax,inhibit the proliferatio
