摘要
利用新型混合型阳离子交换层析介质,通过高压层析柱,对低含量万古霉素粗品进行分离纯化。通过对比实验,确定最佳层析条件:洗杂液为0.5%碳酸氢钠溶液,洗杂液p H值为6.9,洗杂液体积为5 BV,洗脱液碳酸氢钠浓度为1.0%,洗脱乙醇浓度为10%。经此工艺制得的万古霉素纯度>95%,收率>70%。
Separation and purification of low content vancomycin crude by high pressure chromatography column using a new type of mixed cation exchange chromatography medium. The optimum chromatographic conditions were determined by contrast experiment: the concentration, p H value and volume of washing impurity were 0.5% sodium bicarbonate solution,6.9 and 5 BV respectively, the concentration of sodium bicarbonate in eluent was 1%, and the elution concentration of ethanol was 10%. The purity of vancomycin produced by this process is 95%, and the yield is 70%.
