摘要
液泡膜Na^+/H^+逆向转运蛋白(Na^+/H^+antiporter,NHX)基因是植物细胞中一类重要的耐盐基因.本研究在转录组测序的基础上,得到了滨麦液泡膜Na^+/H^+逆向转运蛋白2基因(LmNHX2)的cDNA序列,通过生物学软件对该基因编码的蛋白进行分析和预测,并对其响应盐胁迫的表达模式进行了分析.结果显示,LmNHX2的cDNA序列包含1617bp的开放阅读框,编码538个氨基酸.该基因编码的多肽链疏水性氨基酸占多数,具有典型的跨膜蛋白的结构特点.氨基酸序列比对结果表明,LmNHX2蛋白与小麦TaNHX2,长穗偃麦草Te NHX2及大麦Hv NHX1的氨基酸序列高度相似,四者在进化关系上居于同一分支.荧光定量PCR结果显示,LmNHX2的表达受到盐分胁迫的诱导.
The gene of vacuolar Na^+/H^+ antiporter( NHX) plays an important role in plants salt-tolerance.LmNHX2 cDNA was abtained based on the transcriptome sequencing from Leymus mollis. Bioinformatics analysis for LmNHX2 was carried out by a set of bioinformatics tools,and expression of LmNHX2 under salt stress was analysed. The results showed that LmNHX2 cDNA included an ORF of 1617 bp encoding 538 amino acids.Structure of LmNHX2 protein processed the typical characteristics of transmembrane protein. And the LmNHX2 protein had high similarity in amino acid sequence with TaNHX2,Te NHX2 and Hv NHX1. The results of real-time PCR showed that the expression of LmNHX2 was induced by salt stress.
