摘要
为研究鸡柔嫩艾美耳球虫(Eimeria tenlla)在间接共培养条件下对于宿主细胞凋亡的抑制作用,采用显微镜、分光光度法研究柔嫩艾美耳球虫与马-达氏牛肾细胞(MDBK细胞)间接共培养时对其凋亡的影响。将柔嫩艾美耳球虫纯化的裂殖子与MDBK细胞通过Transwell小室间接共培养;分别在共培养3,6,9,12 h时测量细胞培养体系中的凋亡因子Caspase2,Caspase3,Caspase6,Caspase8,Caspase9的活性。结果表明,测定的共培养组的5种凋亡因子的水平均低于对照组,说明在间接共培养开始到12 h时,MDBK细胞的凋亡受到了抑制,可以推断球虫裂殖子可能会分泌抑制凋亡的物质作用于宿主细胞,而究竟是何种物质或作用机制使得宿主细胞凋亡受到抑制仍有待进一步的研究。
The aim of study is to investigatethe apoptosis inhibition of the Indirect co-culture MDBK cells with Eimeria. The apoptosis inhibited by Indirect co-culturing Eimeria tenUa with MDBK cells were studied by microscopyand spectrophotometric method. The purified E. tenlla and MDBK cells were co-cultured by the transwell chambers. After 3,6,9, and 12 hours the activity of Caspase2,3,6,8, and 9 was analyzed by spectrophotometric method. The results indicated that the level of Caspase from the indirect co-culture group was obviously lower than that of control group, suggesting that the apoptosis of the MDBK cells was inhibited. The system of transwell indirect co-culture was used to study the level of apoptosis indicated by Toxoplasma exocrine antigen(ESA) with Neural stem ceils (NSCs). Similarly the merozoites still inhibited the apoptosis of the MDBK cells without the invasion of host cell before 12 h. We conclude the inhibitony effect on apoptosis of the host cells may be caused by some unknown-substance from the merozoites.
