超临界流体萃取-色谱法提纯鱼油乙酯中EPA-EE和DHA-EE 被引量:3

摘要 基于自主研发的超临界流体萃取-色谱一体装备,将超临界流体精馏技术和超临界流体色谱技术相结合,对乙酯化鱼油原料在超临界CO2中的溶解情况进行了系统研究,经多种单因素考察,探索了二十碳五烯酸乙酯、二十二碳六烯酸乙酯的分离纯化工艺,并探讨其影响因素。结果表明,鱼油乙酯中的EPA-EE和DHA-EE的最佳分离纯化条件:萃取温度梯度40~90℃,萃取压力14 MPa,CO2体积流量100mL/min,鱼油进样速率2 mL/min,以硅胶为色谱柱填料,色谱分离温度40℃,色谱分离压力14 MPa。在此条件下,鱼油乙酯中EPA-EE和DHA-EE质量分数由30%提升到87%。 Extraction and purification processes of EPA-EE and DHA-EE were explored using the supercritical fluid extraction and chromatography equipment,which was developed by the laboratory.The dissolution of ethyl ester of fish oil in supercritical CO2 was investigated by means of supercritical fluid distillation and supercritical fluid chromatography.The purification process of EPA-EE and DHA-EE was explored,and the influencing factors of the experiment were discussed.The results showed that the optimum extraction conditions were as follows:temperature gradient of 40-90 ℃,extraction pressure of 14 MPa,the volume flow of CO2 of 100 mL/min,the flow rate of fish oil of 2 mL/min,silica gel used as column packing,column temperature of 40 ℃,back pressure of 14 MPa.Under the conditions,total concentration of EPA-EE and DHA-EE in fish oil ethyl ester increased from 30%to 87%.
