摘要
目的探讨济阴颗粒治疗绝经综合征的可能作用机制。方法 105只大鼠随机选择10只作为空白对照组,其余95只采用腹腔注射去氧乙烯基环己烯溶液建造绝经综合征大鼠模型,将造模成功60只大鼠随机分为模型组、西药对照组、中药对照组和济阴颗粒低、中、高剂量组,每组10只。造模后空白对照组与模型组给予蒸馏水10ml/(kg·d)灌胃,济阴颗粒低、中、高剂量组分别给予济阴颗粒3.9、7.8、15.6 g济阴颗粒/(kg·d)灌胃,西药对照组给予戊酸雌二醇片0.105 mg/(g·d)灌胃,中药对照组给予坤宝丸10.5g/(kg·d)灌胃,每日灌胃1次,连续30天。免疫组织化学法检测卵巢细胞P53蛋白表达,荧光RT-PCR法检测卵巢细胞凋亡诱导因子(AIF)和核酸内切酶G(Endo-G)mRNA表达。结果与空白对照组比较,模型组卵巢细胞P53蛋白表达及AIF、Endo-G mRNA表达均升高(P<0.01),而济阴颗粒各剂量组均能降低卵巢细胞P53蛋白、AIF mRNA与Endo-G mRNA表达(P<0.01)。济阴颗粒低剂量组下调P53蛋白最显著,而高剂量组下调AIF mRNA作用最显著(P<0.01)。结论济阴颗粒可抑制Caspase非依赖性细胞凋亡通路,从而抑制卵巢细胞凋亡,可能是其治疗绝经综合征的作用机制之一。
Objective To explore the possible mechanism of Jiyin Granule（济阴颗粒） in the treatment of menopause syndrome. Methods Ten rats were randomly selected out of 105 rats as the control group,and the rest 95 rats were used to build rat model of menopause syndrome by intraperitoneal injection of 4-vinylcyclohexene diepoxide solution. Sixty successful modeling rats were randomly divided into a model group,a western medicine control group,a Chinese medicine control group and Jiyin Granule low,middle and high dose groups,with 10 rats in each group.After modeling,the control group and the model group were given 10 ml/（ kg·d） of distilled water by gavage. The Jiyin Granule low,middle and high dose groups were given 3. 9,7. 8,and 15. 6 g/（ kg·d） of Jiyin Granule by gavage,respectively. The western medicine control group was given 0. 105 mg/（ g·d） of Estradiol Valerate Tablets by gavage. The Chinese medicine control group was given 10. 5 g/（kg·d） of Kunbao Pills（坤宝丸） by gavage. All of above treatment were given daily for 30 consecutive days. The expression of p53 protein in ovarian cells was detected by immunohistochemistry. The expression of apoptosis inducing factor（ AIF） and Endonuclease G（ Endo-G） mRNA in ovarian cells were detected by fluorescence RT-PCR. Results Compared with those in the blank control group,the expression of p53 protein and the expression of AIF and Endo-G mRNA in the ovarian cells in the model group were all increased（ P 0. 01）,while the expression of p53 protein,AIF mRNA and Endo-G mRNA in the ovary cells were reduced in the groups of different doses of Jiyin Granule（ P 0. 01）. The down-regulation of p53 protein in the Jiyin Granules low dose group was the most significant,while down-regulation of AIF mRNA in the high dose group was the most significant（ P 0. 01）. Conclusion Jiyin Granule could inhibit the Caspase independent apoptosis pathway and inhibit the apoptosis of ovarian cells,which may be one of its mechanisms in the treatment of me
