摘要
目的研究云南汉族多形性日光疹(PLE)患者的HLA-DPA1、DPB1和DRB1等位基因和单倍型的多态性,探讨多形性日光疹的发病机制。方法采用PCR-SBT法检测云南汉族PLE患者和健康人群HLA-DPA1、DPB1和DRB1基因分布,比较两组间等位基因和单倍型频率,分析其与PLE发病的相关性。结果 HLA-DPA1~*02:02:03、DPB1~*05:01:01、DPB1~*14:01:01,DRB1~*08:03:02等位基因和DPA1~*02:02:03-DPB1~*05:01:01单倍型在病例组中频率明显升高,HLA-DPA1~*02:01:02,DPA1~*02:02:02、DRB1~*03:01:01、DRB1~*11:01:01、DRB1~*12:02:01等位基因和DPA1~*02:02:02-DRB1~*03:01:01单倍型在病例组中频率明显降低,未发现有统计学意义的三位点单倍型。结论 HLA-DPA1、DPB1和DRB1与云南汉族多形性日光疹发病具有相关性。
Objective To study the polymorphism of HLA-DPA1, DPB1 and DRB1 alleles and haplotypes in Han population with polymorphous light eruption and explore its pathogenesis in Yunnan area.Methods PCR-SBT method was applied to detect HLA-DPA1,DPB1 and DRB1 gene distributions in PLE patients and healthy subjects in Yunnan Han population in order to compared the frequencies of allele and haplotype between the two groups and then to analyse its correlation with the incidence of PLE. Results The frequencies of HLA-DPA1*02:02:03,DPB1*05:01:01、DPB1*14:01:01,DRB1*08:03:02 alleles and DPA1*02:02:03-DPB1*05:01:01 haplotype were much higher in the PLE group than the control group, but the frequencies of DPA1*02:01:02, DPA1*02:02:02, DRB1*03:01:01, DRB1*11:01:01, DRB1*12:02:01 alleles and DPA1*02:02:02-DRB1*03:01:01 haplotype were much lower in the PLE group than the control group.There was no statistically significant difference between three locus haplotype.Conclusion HLA-DPA1, DPB1 and DRB1 are related to the incidence of polymorphous light eruption in Yunnan Han population.
