菊糖活化血清补体系统及SC5b-9的纯化

摘要 用菊糖inulin与人血清温育(37℃),1h后血清中补体即大量激活,3h后完全激活;用DEAE-Sephacel层析,蔗糖密度梯度离心和抗IgMprotein A亲和层析法纯化SC5b-9,经免疫电泳及膜攻击复合物抗血清检测呈单一沉淀弧,在SDS-PAGE电泳中显示C5b、C6、C7/C8αγ、C8β、C9、C9_2、S-蛋白的条带,表明SC5b-9已被纯化。 During a study of the effect of inulin on human complement, we found that complements were partially activated after incubation with inulin for one hour and completely activated after incubation for three hours at 37℃. SC5b--9 was purified by DEAE--Sephacel chromatography, linear sucrose density gradi- ents centrifugation and anti--IgM--protein A affinity chromatography. Purified SC5b--9, as assayed by immunoelectrophoresis, was a single fraction and had C5b, C6, C7/C8αγ, C8β, C9, C9_2, and S--protein bands appearing in the 10% SDS--PAGE.
