摘要
目的检测急性心肌梗死患者血浆micro RNA-1(mi R-1)、尿路上皮癌相关1基因(UCA1)的表达水平并探讨其临床意义。方法选择2015年3月~2016年12月于青岛大学附属心血管病医院心内科收治的急性心肌梗死患者67例纳入病例组,男性39例,女性28例,年龄39~76岁,平均(56.3±8.1)岁。并于同期随机选取50例健康体检者为对照组,男性29例,女性21例,年龄42~75岁,平均(54.6±9.2)岁。留取对照组入组后,急性心肌梗死患者入院后不同时段(入院后2 h、8 h、12 h、24 h、7 d、14 d)的血浆标本,采用实时荧光定量PCR检测mi R-1、UCA1的表达。结果病例组患者血浆mi R-1的水平高于对照组,血浆UCA1的水平低于对照组,差异均有统计学意义(P均<0.05)。急性心肌梗死患者入院后,血浆mi R-1水平先呈上升趋势,在24 h达到最高值,随后呈逐渐下降趋势,并且入院后2 h、14 d的血浆mi R-1水平均高于对照组,差异有统计学意义(P均<0.05);血浆UCA1水平先呈逐渐下降趋势,在24 h降到最低值,随后呈逐渐上升趋势,且入院后2 h、14 d时仍低于对照组,差异有统计学意义(P均<0.05)。急性心肌梗死患者血浆mi R-1与UCA1水平呈负相关关系(r=-0.497,P<0.05);入院后2 h、8 h、12 h、24 h、7 d、14 d,血浆mi R-1与UCA1水平均呈负相关(r=-0.498、-0.492、-0.531、-0.596、-0.422、-0.495,P均<0.05)。结论 Micro RNA-1、UCA1在急性心肌梗死患者血浆中异常表达,并且二者呈负相关关系,联合检测可能作为早期辅助诊断急性心肌梗死的重要标志物。
Objective To detect the expressions of plasma microRNA-1 (miR-1) and urothelial carcinoma associated 1 (UCA1) gene in patients with acute myocardial infarction (AMI), and discuss their clinical significance. Methods AMI patients (n=67, male 39, female 28, aged from 39 to 76 and average age=56.3±8.1) were chosen into patient group from the Cardiovascular Hospital affiliated to Qingdao University from Mar. 2015 to Dec. 2016. At the same time, health controls (n=50, male 29, female 21, aged from 42 to 75 and average age=54.6±9.2) were chosen into control group. The plasma samples were collected from AMI patients at different time points (2 h, 8 h, 12 h, 24 h, 7 d and 14 d after hospitalization), and expressions of miR-1 and UCA1 gene were detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR). Results The expression of plasma miR-1 was higher, and expression of UCA1 gene was lower in patients group than those in control group (all P〈0.05). In patients group, the expression of plasma miR-1 showed an ascending trend after hospitalization, reached the highest after 24 h and then showed a descending trend. The expression of plasma miR-1 was higher in patient group than that in control group at time points of 2 h and 14 d (all P〈0.05). The expression of plasma UCA1 gene showed a descending trend, reached the lowest after 24 h and then showed an ascending trend in patients group. The expression of plasma UCA1 gene was lower in patients group than that in control group at time points of 2 h and 14 d (all P〈0.05). The expression of plasma miR-1 was negatively correlated to expression of plasma UCA1 gene in patient group (r=-0.497, P〈0.05). The expression of plasma miR-1 was negatively correlated to expression of plasma UCA1 gene at time points of 2 h (r=-0.498), 8 h (r=-0.492), 12 h (r=-0.531), 24 h (r=-0.596), 7 d (r=-0.422) and 14 d (r=-0.495, all P〈0.05).
