摘要
克隆了壳聚糖酶基因及其自身信号肽,并将其与p MA5载体连接,利用组成型启动子在枯草芽孢杆菌(Bacillus subtilis)中进行转录表达。结果表明,重组壳聚糖酶工程菌在没有诱导物的条件下实现了表达,同时对壳聚糖酶的酶活性进行了研究,最适温度50℃,最适p H 6,为其工业化奠定了很好的基础。
The chitosanase gene and its own signal peptide have been cloned and ligated with pMA5 vector which was tran- scripted by constitutive promoter. This eliminates the need of adding inducers and greatly reduces the cost. The results also showed that the recombinant strain were successfully expressed in the absence of inducers,whereas the control strains didn't produce in the absence of the inducer. In this paper,the enzymatic activity of chitosanase was studied,and the optimum temperature and pH were studied. The optimum temperature was 50℃ and the optimum pH was 6,which laid a good foundation for its industrialization,
