蝉翼藤单体提取化合物对乙醇诱导小鼠肝脏氧化损伤保护作用研究 被引量:1

Effects of Monomeric Compound Extracted from Securidaca on the Protection of Liver Against Oxidative Damage Induced by Ethanol in Mice

摘要 目的探讨蝉翼藤单体提取化合物——甲基阿魏酸对乙醇诱导小鼠肝脏氧化损伤及B淋巴细胞瘤2相关X蛋白(Bax)、B淋巴细胞瘤2(Bcl-2)表达的影响,研究甲基阿魏酸的保护作用。方法 2017年3—6月,选取SPF级雄性成年C57BL/6小鼠60只,采用随机数字表法分为正常组、模型组和甲基阿魏酸低剂量组(5 mg/kg)、甲基阿魏酸中剂量组(10 mg/kg)、甲基阿魏酸高剂量组(20 mg/kg),每组12只。除正常组以外,其余各组每天早上50%乙醇溶液按照0.1 ml/10 g体质量灌胃。每天下午,甲基阿魏酸给药组分别给予相应剂量的甲基阿魏酸灌胃,连续4周。取肝组织苏木素-伊红(HE)染色,观察其病理改变。计算肝脏指数、脾脏指数,生化分析法测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总胆固醇(TC)、三酰甘油(TG)及肝组织过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平,反转录聚合酶链式反应(RT-PCR)和Western blotting法分别检测肝组织中Bax、Bcl-2 mRNA及蛋白的表达。结果与正常组比较,模型组、甲基阿魏酸低、中、高剂量组小鼠肝脏指数、脾脏指数增加,血清ALT、AST、TC、TG水平升高,肝组织CAT、GSH-Px、SOD水平降低,MDA水平升高,肝组织Bax mRNA/Bcl-2 mRNA、Bax/Bcl-2升高(P<0.05);与模型组比较,甲基阿魏酸低、中、高剂量组小鼠肝脏指数、脾脏指数和血清ALT、AST、TC、TG及MDA水平降低,肝组织CAT、GSH-Px、SOD水平升高,肝组织Bax mRNA/Bcl-2 mRNA、Bax/Bcl-2降低(P<0.05)。甲基阿魏酸高剂量组小鼠以上指标改善最明显,差异均有统计学意义(P<0.05)。结论甲基阿魏酸对乙醇诱导的小鼠肝损伤具有保护作用,可有效改善肝功能,减轻肝损伤程度,维持Bax、Bcl-2的平衡,其机制可能与抵抗肝脏氧化损伤以及抑制细胞凋亡有关。 Objective To investigate the protective effect of methyl ferulic acid,a monomeric compound extracted from securidaca based on analyzing its effect on the oxidative damage in liver and the expressions of Bax and Bcl-2 in mice induced by ethanol. Methods From March to June 2017,60 SPF grade male C57 BL/6 mice were selected and randomized into normal group,model group and low-dose( 5 mg/kg) methyl ferulic acid group,middle-dose( 10 mg/kg) methyl ferulic acid group,high-dose( 20 mg/kg) methyl ferulic acid group,12 in each group. Except the normal group,the other groups were given intragastric administration of 50% ethanol solution in a volume of 0. 1 ml/10 g body weight every morning, the low-dose,middle-dose and high-dose methyl ferulic acid groups additionally received intragastric administration of methyl ferulic acid solution in a volume of 5 mg/kg,10 mg/kg,20 mg/kg body weight,respectively every evening,for 4 weeks. Hepatic tissues of the mice were taken out and stained with HE,and their pathological changes were observed. The liver index and splenic index were calculated. Biochemical analyses of serum ALT,AST,TC, TG, catalase( CAT), glutathione peroxidase( GSH-Px),superoxide dismutase( SOD),malondialdehyde( MDA) levels were performed. The expressions of Bax,Bcl-2 mRNA in liver tissues were detected by reverse transcription-polymerase chain reaction( RT-PCR). The expressions of Bax and Bcl-2 in liver tissues were detected by western blotting. Results Increased values of liver index,splenic index,ALT,AST,TC,TG and MDA,elevated expressions of Bax/Bcl-2 mRNA and Bax/Bcl-2,and decreased values of CAT,GSH-Px,SOD were found in the model group,low-dose,middle-dose and high-dose methyl ferulic acid groups rather than the normal group( P <0. 05). The model group had higher values of liver index,splenic index,ALT,AST,TC,TG,MDA,increased expressions of Bax/Bcl-2 mRNA and Bax/Bcl-2 but lower values of CAT,GSH-Px and SOD than the low-dose,middle-dose and high-dose methyl ferulic acid groups( P < 0. 05).
