多重实时荧光PCR检测食品中致泻性大肠埃希菌 被引量:3

Detection of diarrhoea Escherichia coli in food by multiple real-time fluorescence PCR

摘要 目的快速诊断鉴定致泻性大肠埃希菌及其毒力基因和型别情况,为疾病预防控制提供快速、准确的实验室依据。方法依据《GB 4789.6-2016食品微生物学检验致泻性大肠埃希菌检验》和《2017年国家食品污染物和有害因素风险监测工作手册》,采用多重实时荧光PCR技术,根据扩增曲线的荧光信号类型确定待测目标菌是否含致泻性大肠埃希菌,并判断其致病型别。结果于2017年上半年,按国家食源性致病菌监测计划,抽检42份市售生畜肉、水果、蔬菜食品,结果检出5份样品含致泻性大肠埃希菌,5株菌均为肠聚集性大肠埃希菌(EAEC),其中4株毒素类型是uid A、ast A,另1株毒素类型是uid A、agg R+pic。结论在食品微生物及致病因子监测工作中,应用PCR技术深入分子水平检测致泻性大肠埃希菌,有助于建立致泻性大肠埃希菌的快速准确检验机制,有效控制传染源,为今后细菌鉴定工作提供参考。 Objective With the rapid diagnosis and identification of diarrheogenic Escherichia coli and its virulence genes and types, to provide a rapid and accurate laboratory basis for disease prevention and control. Methods On the basis of "GB 4789.6-2016 Food Microbiology Test Diarrheogenic Escherichia Coli Inspection" and "2017 National Food Contamination and Harmful Factors in Risk Monitoring Handbook",to determine whether the target bacteria containing diarrheagenic Escherichia coli and it's pathogenic type according to the fluorescent signal of amplification curve with multiplex real time fluorescence PCR technology. Results In the first half of 2017, according to the national foodborne pathogenic bacteria monitoring plan, 42 commercially available raw meat, fruits, vegetables, food were checked. 5 samples of diarrheogenic Escherichia coli were detected out, and all 5 strains were Enterotoxigenic Escherichia coli(EAEC), which included 4 strains of uid A, ast A and 1 strain of uid A, agg R +pic. Conclusion In the monitoring of food microbiology and pathogenic factor, to use the detection of diarrheogenic Escherichia coli by PCR technique at molecular level, which is helpful to establish a rapid and accurate detection mechanism of diarrheogenic Escherichia coli, and to effectively control the source of infection, in order to provide reference for bacterial identification in the future.
