摘要
目的测定芒柄花素的平衡溶解度和油水分配系数,为设计芒柄花素新剂型提供理论基础与依据。方法采用高效液相色谱(HPLC)法测定芒柄花素在水、不同缓冲盐溶液中的质量浓度,并运用摇瓶法测定芒柄花素在正辛醇-水、缓冲液体系中的表观油水分配系数。结果 37℃下芒柄花素在水中的平衡溶解度为2.079±0.036μg·mL^(-1),随着pH值增大,芒柄花素的平衡溶解度增大。芒柄花素在正辛醇-水体系中的表观油水分配系数lg P_(app)=2.79±0.03,随着缓冲盐溶液的pH值增大,芒柄花素在正辛醇-缓冲盐溶液体系中的油水分配系数减小。结论建立的方法可以准确测定芒柄花素的平衡溶解度和表观油水分配系数,科学合理地预测药物在体内的吸收与分布情况。
Objective To determine the equilibrium solubility and the apparent oil/water partition coefficient of formononetin for designing new formulations. Methods The mass concentration of formononetin in water or phosphate buffer with different pH was detected by HPLC, and the shake flask method was used to determine the apparent partition coefficient of formononetin in n-octanol-water-buffer solutions of different pH. Result The equilibrium solubility of formononetin was 2.079 ±0.036 μg·mL^(-1) in water at 37 ℃ and increased with the pH of buffer solution increasing. lg P_(app) of formononetin was 2.79 ±0.03 in n-octanol-water system and decreased with the pH of buffer solution increasing. Conclusion The methods founded in this research can be used to determine accurately the equilibrium solubility and apparent partition coefficient of formononetin, and predict the in vivo absorption and distribution of formononetin.
