地锦草对Hela细胞增殖与凋亡的影响 被引量:8

摘要 目的:探讨中药地锦草对Hela细胞增殖和凋亡作用的影响。方法:以人宫颈癌Hela细胞株为研究对象,采用MTT法检测地锦草对Hela细胞增殖抑制作用的影响,AO/EB染色法荧光倒置显微镜观察药物作用后凋亡小体的形成,Annexin V-FITC流式细胞术(FCM)检测细胞凋亡变化情况。结果:地锦草能够抑制Hela细胞的体外增殖,抑制程度随药物浓度50μg·m L^(-1)升高到200μg·m L^(-1),抑制率也由30.4%增大到61.1%,显示了良好的浓度依存关系(P<0.05);AO/EB染色法显示药物作用后凋亡小体形成,FCM法结果显示随着药物作用质量浓度由0增高到200μg·m L^(-1),细胞凋亡率也由0.76%升高到57.17%(P<0.05)。结论:地锦草能够抑制Hela细胞增殖。其作用可能是通过诱导Hela细胞凋亡和分化实现的。 Objective:To investigate the effects of Euphorbia humifusa on proliferation,apoptosis of Hela cervical cancer cells.Methods:The Hela cell line was used for the study.The inhibition of E.humifusa on the proliferation of Hela cells was assayed by MTT.Apoptotic cells was determined using the AO/EB staining,and cell apoptosis was detected by flow cytometry (FCM).Results:E.humifusa could inhibit the growth of Hela cell,the inhibitory rate increased with the drug concentration by the 50 μg·mL-1 to 200 μg·mL-1,inhibition rate also increased from 30.4% to 61.1% in concentration dependent manners(P〈0.05).FCM revealed that the percentage of cells that underwent apoptosis mounts from 0.76% to 57.17% as the treated concentration was elevated from 0 to 200 μg·mL^-1.Conclusion:E.humifusa can efficiently inhibit the proliferation of Hela cells,maybe by the way of leading to its apoptosis.
