摘要
目的:探讨温阳化饮方通过调节基质金属蛋白酶9(MMP-9)的活性来干预支气管哮喘寒饮蕴肺证大鼠气道重塑的机制。方法:采用卵蛋白致敏,加冰箱冷冻、冰水饲养、水中游泳等方法,建立支气管哮喘寒饮蕴肺病证结合大鼠模型,分别用1,25-(OH)2D3、小青龙汤、温阳化饮方、六君子汤进行干预,观察大鼠的一般行为、肺功能的变化及肺组织中MMP-9的活性。结果:造模较为成功,模型组大鼠的一般行为学表现与支气管哮喘寒饮蕴肺证的临床表现基本相符,肺功能及MMP-9的变化也证实了造模较为成功,在实验即将结束时,其余各组较正常组大鼠的吸气阻力(Ri)、呼气阻力(Re)均增大,肺通气顺应性(Cldyn)降低,模型组大鼠肺组织中MMP-9的活性明显高于治疗组;而治疗组的4种药物对大鼠的行为、肺功能及MMP-9的表达都有不同程度的改善,其中温阳化饮方疗效总体最佳,温阳化饮方对大鼠的症状和体征改善情况最好,对肺功能的改善情况也最佳(P<0.05),在减少MMP-9的活性及m RNA表达方面也表现最强(P<0.01)。结论:温阳化饮方能够通过减少MMP-9的活性来抑制支气管哮喘寒饮蕴肺证气道重塑的作用,减轻支气管哮喘寒饮蕴肺证的症状。
Objectives:To investigate the mechanism on airway remodeling of cold fluid accumulation in lung of bronchial asthma intervened with Wenyang Huayin Decoction by adjusting MMP-9 activity. Methods: The rat model of cold fluid accumulation in lung syndrome of bronchial asthma was established by methods of ovalbumin sensitization, refrigeration, raising in ice water, swimming in warm water. Then, 1,25-(OH)2D3, Xiaoqinglong Decoction, Wenyang Huayin Decoction, and Liujunzi Decoction were adopted in experiment respectively to observe the behaviors of rats, changes of lung function and MMP-9 activity in lung tissue. Results: The rat model is successfully made because the behaviors of rats of experiment group are consistant with clinical manifestations of asthma with syndrome of cold-fluid accumulation in lung, which is also confirmed by changes of lung function and MMP-9, at the end of the experiment, in other groups, the resistant inspiration (Ri), resistant expiration (Re) of rats are increased, and compliant lung dynamis (Cldyn) of rats are lower than those in normal rats, the levels of MMP-9 activity in lung tissue is significantly higher than the treatment groups. The four above mentioned formulas can improve the syndrome, such as the behaviors of rats, changes of lung function and the levels of MMP-9 activity, among which formula of wanning yang and resolving rluid retention is the most effective in terms of efficacy. Foumula of warming yang and resolving rluid retention on rats to improve the symptoms are the best, the improvement of lung function is also the best (P〈0.05), and it also shows the strongest activity in reducing MMP-9 activity and mRNA expression (P〈0.01). Conclusion: Wenyang Huayin Decoction can inhibit airway remodeling of cold fluid accumulation in lung syndrome of bronchial asthma by inhibiting the activity of MMP-9. Alleviates symptoms of cold fluid accumulation in lung syndrome of bronchial asthma.
