螺旋藻制备高活性小肽的研究

摘要 以螺旋藻为原料,使用毕赤酵母发酵所产生的蛋白酶对其水解制备了高活性小肽。对酶解工艺条件进行研究,根据正交试验设计原理,在单因素试验的基础上,以DH和TCA-SNI为指标,对影响螺旋藻水解的各种影响因素:温度、pH、料水比、酶底比和时间进行了系统研究,得到了最佳工艺条件为:料水比1∶8、酶底比700U/g、酶解温度55℃、pH值8.5,酶解时间3h。在最佳条件下制备螺旋藻活性肽的水解度(DH)为11.82%,三氯乙酸-可溶性氮指数(TCA-SNI)为55.13%。利用凝胶过滤色谱法对酶解产物进行分子量的测定,结果显示:分子量小于1000Da的寡肽占55.02%,小肽分子量分布主要集中在260Da附近,约2个氨基酸残基,含量为29.07%。制备的螺旋藻肽粗蛋白含量为62.52%,总磷含量为1.00%,钙含量为0.13%,铁含量为803mg/kg。 The preparation of highly active peptides from Spirulina mud by the enzyme which produced by plchiater- mentation was studied. The orthogonal experiment was used to study the hydrolysis conditions. The influence factors, including temperature,pH,the ratio of material to water, enzyme dosage and hydrolysis time,were investigated according to DH and TCA-SNI. The optimum hydrolysis conditions were as follows., the ratio of material to water 1 ~ 8, enzyme dosage 700 U/g, hydrolysis temperature 55℃, pH 8.5, hydrolysis time 3 h. Under the optimal conditions, the DH of the hydrolysates reached 11.8% and the TCA-SNI of the hydrolysates reached 55.13%. The molecular weight of enzymolysis product was determined by Gel filtration chromatography. The result showed that the oligopeptide which the molecular weight was less than 1000 Da reached 55.02%. And the small peptide mainly with 260 Da molecular weight composed by 2 amino acids reached 29.07 %. The hydrolysates had abundant nutrients, protein 62. 52% ,total phosphorus 1.00% ,calcium 0.13% and iron 803 mg/kg.
