摘要
为开发以麝香草酚为主要成分的新型抑菌剂。采用微量肉汤稀释法测定麝香草酚对单增李斯特菌的最小抑制浓度和最小致死浓度分别为156.25μg/ml和312.50μg/ml。通过扫描电镜观察发现,经过麝香草酚处理后单增李斯特菌细胞出现穿孔式膜损伤。进一步研究结果表明,麝香草酚分别处理1 h、2 h和3 h后,单增李斯特菌胞外的碱性磷酸酶、葡萄糖和核酸含量显著高于对照。SDS-PAGE电泳分析结果显示,麝香草酚处理6 h后单增李斯特菌胞内蛋白含量降低,说明麝香草酚处理使得单增李斯特菌细胞通透性增加,导致细胞内的物质流失到细胞外。碘化丙啶(PI)染色结合倒置荧光显微镜观察结果显示,麝香草酚处理造成了单增李斯特菌细胞膜的破损,并最终导致菌体细胞死亡。
The inhibitory effect of thymol against Listeria monocytogenes was investigated in this study. The minimum inhibitory concentration and minimal bactericidal concentration of thymol were 156.25 μg/ml and 312.50 μg/ml respectively determined by micro broth dilution method. Scanning electron microscopic observation revealed that the surface structure of thymol-treated L. monocytogenes was damaged. Extracellular contents of alkaline phosphatase,glucose and DNA from L.monocytogenes were significantly higher than those of control after treated with thymol for 1-3 h. The SDS-PAGE pattern of intracellular soluble proteins of L. monocytogenes was shallow after thymol treatment for 6 h,suggesting that thymol caused the increase of membrane permeability and the leakage of intracellular substances from L. monocytogenes. Observation under inverted fluorescence microscope after PI stain indicated that thymol induced the membrane damage and cell death of L.monocytogenes.
