摘要
目的探讨Orai1和STIM1蛋白在结肠腺癌中的表达及其临床意义。方法采用免疫组化法检测80例结肠腺癌及50例正常结肠黏膜中Orai1和STIM1蛋白的表达。分析两者表达与结肠腺癌临床病理特征及预后的关系。结果结肠腺癌组织中Orai1和STIM1表达水平均显著高于正常结肠黏膜(P均<0.05);Orai1和STIM1表达与肿瘤TNM分期及淋巴结转移有关(P<0.05),与患者性别、年龄、分化程度无关(P>0.05)。结肠腺癌中Orai1与STIM1蛋白表达呈正相关(P=0.001,rs=0.349)。单因素生存分析显示,Orai1蛋白、STIM1蛋白、TNM分期及淋巴结转移与结肠腺癌患者预后相关(P<0.05)。结论 Orai1和STIM1可能具有协同作用,共同促进结肠癌的发展,有望成为结肠癌诊断、治疗以及判断预后的生物学指标。
Purpose To investigate the expression of Orail and STIM1 in colon cancer and its clinical significance. Meth- ods Immnohistochemistry was used to detect Orail and STIM1 protein expression level in 80 cases of colon cancer and 50 cases of normal colon epithelium. The relationships between Oral1 and STIM1 expression and prognosis were statistically analyzed. Re- sult The expression levels of Orail and STIM1 in colon cancer were significantly higher than that in normal colon epithelium（P 〈 O. 05 ）. Positive expression of Orail and STIM1 was correlated with the TNM stage and lymph node metastasis（ P 〈 O. 05 ）, but not correlated with gender, age, and differentiation（ P 〉 0. 05 ）.There was a significant positive correlation between Orail and STIM 1 expression （ P = 0. 001, rs = 0. 349 ）. Univariate analysis showed that the expression of Orail protein, STIM1 protein, TNM stage and lymph node metastasis were significant prognostic factors for colon cancer patients. Conclusion In colon cancer, both Orail and STIM1 proteins may have synergetic functions and promote the development of the tumor, and could be used as a novel biological indicator of anticancer therapy and prognosis.
