病毒性心肌炎患儿外周血miR-146b及miR-155表达量与Treg免疫、心肌损伤的相关性 被引量:15

摘要 目的:研究病毒性心肌炎患儿外周血miR-146b及miR-155表达量与Treg免疫、心肌损伤的相关性。方法:选择在本院诊断为病毒性心肌炎的患儿作为研究的VMC组,同期健康体检的儿童作为研究的对照组。采集外周血并测定miR-146b、miR-155及免疫转录因子的表达量,采集血清并测定免疫细胞因子及心肌损伤指标的含量。结果:VMC组患儿外周血中miR-146b、miR-155的表达量显著高于对照组;VMC组患儿外周血中Foxp3的mRNA表达量以及血清中IL-10、TGF-β的含量显著低于对照组且与miR-146b、miR-155的表达量呈负相关,外周血中ROR-γt的mRNA表达量以及血清中IL-17、IL-21、CK-MB、cTnI、GrB、sFasL、caspase-3的含量显著高于对照组且与miR-146b、miR-155的表达量呈正相关,结论:病毒性心肌炎患儿外周血中高表达的miR-146b及miR-155能够抑制Treg免疫并加重心肌损伤。 Objective:To study the correlation of peripheral blood mir-146 band mir-155 expression with Treg immunity and myocardial injury in children with viral myocarditis.Methods:The children who were diagnosed with viral myocarditis in Wuhan Children's Hospital between June 2014 and February 2017 were selected as the VMC group of the study,and the children who received physical examination during the same period were selected as the control group of the study.Peripheral blood was collected to determine the expression of mir-146 b,mir-155 and immune transcription factors,and the serum was collected to determine the contents of immune cytokines and myocardial injury indexes.Results:Peripheral blood mir-146 band mir-155 expression of VMC group were significantly higher than those of control group;peripheral blood Foxp3 mRNA expression as well as serum IL-10 and TGF-βcontents of VMC group was significantly lower than those of control group and negatively correlated with mir-146 band mir-155 expression while peripheral blood ROR-γt mRNA expression as well as serum IL-17,IL-21,CK-MB,cTnI,GrB,sFasL and caspase-3 contents was significantly higher than those of control group and positively correlated with mir-146 band mir-155 expression.Conclusion:Highly expressed mir-146 band mir-155 in peripheral blood of children with viral myocarditis can inhibit Treg immunity and increase myocardial injury.
