高氧肺损伤大鼠肺泡内TNF-α IL-1β和IL-6的含量变化研究 被引量:3

摘要 目的探讨细胞因子TNF-α、IL-1β和IL-6在高氧肺损伤发病过程中的含量变化。方法将新生SD大鼠随机分为对照组和高氧组,在实验的第1,3,7天每组随机处死20只,其中10只取肺组织测定肺湿/干重(W/D)比值,另10只行支气管肺泡灌洗,ELISA法测定肺泡灌洗液(BALF)中TNF-α、IL-1β、IL-6含量。结果与对照组相比,高氧组大鼠W/D比值、BALF中TNF-α和IL-1β水平在第1,3,7天均显著升高(P<0.05),高氧组TNF-α水平于第3天较第1,7天高(P<0.01);IL-6水平则在高氧暴露后第3天和第7天明显增加(P<0.05)。结论细胞因子TNF-α、IL-1β和IL-6均参与了高氧肺损伤发病过程。 Objective To investigate the variation of tumor necrosis factor-α(TNF-α)、interleukin(IL)-1βand IL-6 contents in the neonatal hyperoxic lung injury. Methods SD newborn rats were randomLy divided into controlgroup and hyperoxia (95% oxygen)group. After 1,3,7 d of continuous exposure to high concentration oxygen,thewet/dry weight ratio (W/D)of the lung were determined ,the levels of TNF-α、IL-1β and IL-6 in bronchoalveolarlavage fluid (BALF)were measured by the enzyme-linked immunosorbent assay methods. Results Afer 1,3,7d,compared with control group,the wet/dry weight ratio (W/D)of the lung ,levels of TNF-α and IL-1β in BALF ofhyperoxia group obviously increased(P〈0.05).But the level of TNF-α on 3d in hyperoxia group was gradually higherthan that of 1、7d(P〈0.01).The content of IL-6 in BALF of neonatal SD rats on 3、7d in hyperoxia group were higher thanthose of control groups. Conclusion The contents of TNF-α、IL-1β and IL-6 may participate in the pathologicalprogress of hyperoxic lung injury.
